摘 要:目的研究探讨“土茯苓-薏苡仁”药对治疗骨性关节炎的关键靶点以及作用机制。方法通过中药系统药理学分析平台(TCMSP)检索并筛选“土茯苓-薏苡仁”主要有效活性成分及其作用靶点,通过GeneCards数据库、OMIM数据库、PharmGkb数据库、TTD数据库、DrugBank数据库检索收集“骨性关节炎”的相关靶点,制作Venn图获取“土茯苓-薏苡仁”药对与骨性关节炎的共同靶点,运用Cytoscape软件构建“中药活性成分-骨性关节炎-靶点-疾病”网络,利用STRING数据库构建蛋白相互作用网络。最后运用R语言进行基因本体论(GO)功能富集分析和京都基因与基因组百科全书(KEGG)通路富集分析。结果中药“土茯苓-薏苡仁”对骨性关节炎有治疗作用的活性成分共19个(槲皮素等);发挥治疗作用的关键靶点共81个(丝氨酸/苏氨酸蛋白激酶1等);发挥疗效干预的生物学进程主要有对脂多糖的反应、对细胞外刺激的反应等;发挥重要作用的生物学通路主要有脂质和动脉粥样硬化通路、白细胞介素-17(IL-17)信号通路、肿瘤坏死因子(TNF)信号通路等。结论“土茯苓-薏苡仁”药对能够通过多成分、多靶点、多通路治疗骨性关节炎,为后续进一步研究和开展实验工作提供了思路和理论依据。Objective To explore the key target and mechanism of"Smilacis Glabrae Rhizoma-Coicis Semen"for the treatment of osteoarthritis.Methods Traditional Chinese Medicine System Pharmacology Analysis Platform was used to search and screen the main active components and targets of Smilacis Glabrae Rhizoma-Coicis Semen.The related targets of"osteoarthritis"were collected by GeneCards database,OMIM database,PharmGkb database,TTD database and DrugBank database.Venn diagram was made to obtain the common target of"Smilacis Glabrae Rhizoma-Coicis Semen"and osteoarthritis diease.Cytoscape software was used to construct the network of"Active Component-Osteoarthritis-Target-Disease".STRING platform was used to construct the protein-protein interaction network.Gene Ontology(GO)and Kyoto Encyclopedia of Genes and Genomes(KEGG)were carried out to perform functional enrichment analysis with R language.Results There were 19 active components(Quercetin,etc.)in the treatment of osteoarthritis,and 81 key targets in Chinese medcine"Smilacis Glabrae Rhizoma-Coicis Semen".Response to lipopolysaccharide and response to extracellular stimulus were the main biological processes for the occurrence and progression of osteoarthritis disease.The key pathways include lipid and atherosclerosis,interleukin-17(IL-17)signaling pathway,and tumor necrosis factor(TNF)signaling pathway,etc.Conclusion The effect of Chinese medicine on osteoarthritis may be realized through multi-components,multi-targets and multi-channels.
