摘  要:本研究以试验前期筛选到的一株野生产木质纤维素酶活性的食用菌为材料,根据形态学与rDNA-ITS序列对该食用菌进行鉴定。以园林废弃物粉为主要原料,采用单因素试验及正交试验对固体产酶条件进行优化。结果表明:试验食用菌菌株为毛头鬼伞Coprinus comatus。单因素试验证明:氮源、初始pH、温度和接种量为降解木质纤维素的显著影响因子,正交试验确定毛头鬼伞产羧甲基纤维素酶活性最优条件为:pH为7、温度25℃、接种量10%、碳氮比为15:1。该研究结果为园林废弃物木质纤维素的降解提供了良好的菌种资源,同时也为后续园林废弃物木质纤维素降解方面的研究积累了相关数据。In this study,a wild lignocellulase producing edible fungus selected in the early stage of the experiment was identified according to the combination of morphology and ITS biology.Using garden waste powder as the main raw material,solid single factor test and orthogonal test were used to optimize the condition of enzyme production.The results showed that the strain was preliminarily identified as Coprinus comatus by morphological and molecular biological characteristics.Single factor test showed that nitrogen source,initial pH,temperature and inoculum size were significant influencing factors for lignocellulose degradation.Orthogonal test determined that the optimal conditions for carboxymethyl cellulase production were pH 7,temperature 25℃,inoculum size 10%and C/N ratio 15:1.The research results provided a good strain resource for the degradation of lignocellulose from garden waste,and also accumulate relevant data for the subsequent research on lignocellulose from garden waste.
