摘 要:对灵芝多糖的微波辅助水提取、碱提取工艺进行探究。以多糖得率为指标,采用响应面分析法对灵芝多糖的微波辅助水提取和碱提取工艺进行优化。水提取的最佳工艺条件为:微波功率325 W,料液质量比1∶35、提取时间24 min;碱提取的最佳工艺条件:NaOH质量分数5.4%,料液质量比1∶35、提取温度40℃、提取时间75 min。利用优化后的工艺对沪农灵芝和龙芝2号的子实体进行多糖提取,水溶性多糖(GLP1)得率均大于1%,碱溶性多糖(GLP2)得率均大于5%。多糖的红外谱图显示:沪农灵芝的GLP1、GLP2和龙芝2号的GLP1、GLP2均有可能是氨基多糖;沪农灵芝的GLP2和龙芝2号的GLP2中存在吡喃糖环、甘露糖苷。优化后的工艺能显著提高灵芝水溶性、碱溶性多糖产量,不同灵芝材料中水溶性多糖、碱溶性多糖具有差异性。To explore the process of microwave-assisted water extraction and alkali extraction of Ganoderma lucidum.With the yield of polysaccharide as the index,the extraction process were analyzed by response sarface methodology.The optimal conditions of water extraction were as follows:microwave power of 325 W,solvent-material ratio of 1∶35,extraction time of 24 min.The optimal conditions of alkali extraction were as follows:the quality fraction of NaOH liquor of 5.4%,solvent-material ratio of 1∶35,extraction temperature of 40℃,extraction time of 75 min.Under these conditions,the extraction yield of water-soluble polysaccharides(GLP1)was more than 1%,and the extraction yield of alkali-soluble polysaccharide(GLP2)was more than 5%.According to the infrared spectrum of each polysaccharide component,pyranose rings and mannosides might exist in GLP2 of Ganoderma lucidum SH and GLP2 of Longzhi No.2.The optimized process could significantly increase the yields of water-soluble polysaccharides and alkali-soluble polysaccharides of Ganoderma lucidum.There were differences in water-soluble polysaccharides and alkali-soluble polysaccharides in different ganoderma lucidum materials.
