摘 要:目的:考察并优化纤维素酶提取南靖金线莲多糖的工艺参数,建立生物酶提取金线莲多糖的方法。方法:采用酶提醇沉法提取、Savege法纯化金线莲多糖,并应用紫外-可见分光光度法检测金线莲多糖含量。通过试验考察酶添加量、酶解时间、PH缓冲液、酶解温度、料液比等对纤维素酶提取金线莲多糖效果的影响,选取各因素的最优试验范围,对纤维素酶提取金线莲多糖工艺进行优化。结果:金线莲多糖纤维素酶提取最佳条件为:酶添加量为5.0mL、酶解时间180min、PH6.6缓冲液、酶解温度65℃、料液比1:30。在最优提取工艺条件下,金线莲多糖提取率为20.93%。结论:本方法操作简单,快速,重复性好,可为生物酶提取金线莲多糖成分有效利用提供依据。Objective:The technological parameters of cellulase extraction of Nanjing Anoectochilus roxburghii polysaccharide were optimized,and the method of enzymatic extraction was established.Methods:The polysaccharide of Nanjing Anoectochilus roxburghii was extracted by enzyme extraction and alcohol-precipitation method,purified by Savege method,and its content was detected by Uv-vis.The effects of enzyme dosage,enzymolysis time,PH buffer,enzymolysis temperature and solid-liquid ratio on the extraction of efficiency of Anoectochilus roxburghi polysaccharide by cellulase were investigated,and the optimum experimental range of each factor was selected to optimize the extraction process of Anoectochilus roxburghii polysaccharide by cellulase.Results:The optimum conditions for cellulase extraction of polysaccharides from Anoectochilus roxburghii were as follows:5mL enzyme dosage,180min enzymolysis time,PH6.6 buffer,65℃enzymolysis temperature,and solid-liquid ratio of 1:30.Under the optimum extraction conditions,the extraction rate of polysaccharides from Anoectochilus roxburghii was 20.93%.Conclusion:The method is simple,rapid and reproducible,which can provide a basis for the effective utilization of polysaccharide components extracted from Anoectochilus roxburghii by biological enzyme.
