摘  要:目的 研究失神七同系物2(SIAH2)和血管内皮生长因子D(VEGF-D)在非小细胞肺癌(NSCLC)中的表达并探讨两者之间的相关性。方法 采用免疫组织化学分析SIAH2和VEGF-D蛋白在NSCLC中的表达情况,并分析二者与NSCLC的临床病理特征及淋巴管密度(LVD)之间的关系,Log-rank法分析SIAH2表达与患者生存预后的关系,采用SIAH2 siRNA沉默SIAH2表达,同时应用westernblot和RT-PCR检测SIAH2和VEGF-D蛋白和mRNA表达情况。结果 免疫组化染色显示SIAH2和VEGF-D在NSCLC组织中高表达,SIAH2蛋白表达与NSCLC高TNM分期(χ^(2)=7.236,P=0.007)和淋巴结转移(χ^(2)=14.911,P<0.01)有关。SIAH2与VEGF-D的表达成正相关(r=0.499,P<0.01)。SIAH2和VEGF-D的阳性表达组中LVD高于SIAH2和VEGF-D的阴性表达组(P<0.05)。SIAH2蛋白高表达与NSCLC患者预后不良相关(χ^(2)=5.748,P=0.019)。SIAH2的表达和淋巴结转移是该组患者生存时间的独立预测因素。沉默SIAH2表达后SIAH2和VEGF-D表达水平降低(P<0.05)。结论 SIAH2表达与NSCLC患者不良预后相关,可能在NSCLC发生中发挥一定作用,同时SIAH2可能与VEGF-D协同作用影响NSCLC的淋巴浸润和淋巴结转移。Objective This paper aims to study the expression of SIAH2(SIAH2) and VEGF-D in non-small cell lung cancer(NSCLC) and explore the correlation between them. Methods Immunohistochemistry was used to analyze the expression of SIAH2 and VEGF-D proteins in NSCLC, and the correlation between them and the clinicopathological features and lymphatic vessel density(LVD) of NSCLC was analyzed. Log-rank method was used to analyze the relationship between SIAH2 expression and survival prognosis of patients. SIAH2 siRNA was used to silence the expression of SIAH2, and the protein and mRNA expression of SIAH2 and VEGF-D were detected by Western blot and RT-PCR. Results Immunohistochemical staining showed that SIAH2 and VEGF-D were highly expressed in NSCLC tissues, and the level of SIAH2 protein expression was associated with high TNM stage(χ^(2)=7.236, P=0.007) and lymph node metastasis(χ^(2)=14.911, P<0.01). The expression of SIAH2 was positively correlated with the expression of VEGF-D(r=0.499, P<0.01). The LVD in positive-SIAH2 and positive-VEGF-D groups was higher than that in negative-SIAH2 and negative-VEGF-D groups(P<0.05). High expression of SIAH2 protein was associated with poor prognosis in NSCLC patients(χ^(2)=5.748, P=0.019). SIAH2 expression and lymph node metastasis were independent predictors of survival in this group. Moreover, the levels of SIAH2 and VEGF-D were decreased in the cells transfected with SIAH2 siRNA(P<0.05). Conclusion SIAH2 expression is associated with poor prognosis of NSCLC patients and may play a role in the development of NSCLC. Meanwhile, SIAH2 maycooperate with VEGF-D to influence lymphatic infiltration and lymph node metastasis in NSCLC.
