摘 要:目的探讨子宫内膜癌(EC)组织中长链非编码RNA(LncRNA)核富含丰富的转录本1(NEAT1)、微小RNA(miR)-29a的表达及临床意义。方法选取2017年7月至2022年6月在郑州人民医院诊治的EC患者45例,检测癌组织和癌旁组织LncRNA NEAT1、miR-29a表达,并分析癌组织中LncRNA NEAT1、miR-29a表达与临床病理特征的关系,以及LncRNA NEAT1、miR-29a表达对EC的诊断价值。结果EC患者癌组织LncRNA NEAT1表达明显高于癌旁组织(t=7.951,P<0.001),miR-29a表达与癌旁组织比较明显较低(t=5.616,P<0.001)。肿瘤大小≥2cm、临床分期Ⅲ~Ⅳ期EC患者癌组织中LncRNA NEAT1表达高于肿瘤大小<2cm、临床分期Ⅰ~Ⅱ期EC(t=2.251,P=0.030;t=2.273,P=0.028);临床分期Ⅰ~Ⅱ期EC患者癌组织中miR-29a表达高于临床分期Ⅲ~Ⅳ期患者(t=3.091,P=0.003)。ROC曲线分析得出,LncRNA NEAT1、miR-29a、两者联合诊断测EC的AUC分别为0.861、0.700、0.934。在最佳临界值点对应的敏感性、特异性分别为:LncRNA NEAT1为62.2%、100.0%,miR-29a为100.0%、48.9%,两者联合为100.0%、75.6%。结论EC癌组织LncRNA NEAT1呈高表达,miR-29a呈低表达,均和临床分期有关,且前者还与肿瘤大小有关,LncRNA NEAT1、miR-29a联合可为诊断EC提供参考。Objective To investigate the expression and clinical significance of long non-coding RNA(LncRNA)nuclear abundant transcript 1(NEAT1)and microRNA(miR)-29a in endometrial cancer tissue.Methods A total of 45 EC patients who were diagnosed and treated in Zhengzhou people’s Hospital from July 2017 to June 2022 were selected to detect the expression of LncRNA NEAT1 and miR-29a in cancer tissue and adjacent tissue,and analyze the relationship between the expression of LncRNA NEAT1 and miR-29a in cancer tissue and clinical Pathological relationship,and the diagnostic value of tissue LncRNA NEAT1 and miR-29a expression in EC.Results The expression of LncRNA NEAT1 in the carcinoma tissue of EC patients was significantly higher than that in the adjacent tissue(t=7.951,P<0.001),and the expression of miR-29a was significantly lower than that in the adjacent tissue(t=5.616,P<0.001).The expression of LncRNA NEAT1 in cancer tissue of EC patients with tumor size≥2cm and clinical stage Ⅲ-Ⅳ was significantly higher than that of EC patients with tumor size<2cm and clinical stage Ⅰ-Ⅱ(t=2.251,P=0.030;t=2.273,P=0.028).The expression of miR-29a in tissue was significantly higher than that in patients with clinical stage Ⅲ-Ⅳ EC(t=3.091,P=0.003).The ROC curve analysis showed that the AUC of LncRNA NEAT1,miR-29a,and their combined diagnostic tests for EC were 0.861,0.700,and 0.934,respectively.The corresponding sensitivity and specificity at the optimal cut-off point were:62.2% and 100.0% for LncRNA NEAT1,100.0% and 48.9% for miR-29a,and 100.0% and 75.6% for the combination of the two.Conclusion The high expression of LncRNA NEAT1 and the low expression of miR-29a in EC cancer tissue are related to clinical stage,and LncRNA NEAT1 in EC cancer tissue is related to tumor size.The combination of LncRNA NEAT1 and miR-29a can provide a reference for the diagnosis of EC.
