摘 要:目的观察高脂饮食诱导的不同肥胖程度大鼠皮下脂肪的分化状态,探讨不同肥胖程度对大鼠皮下脂肪分化BMP4/PPARγ信号通路的影响。方法SD大鼠随机分为普通饲料(NC)组,高脂饲料组,高脂饲料组建立不同程度的肥胖模型,分别为超重(ML)组、轻度肥胖(MN)组和中度肥胖(MS)组,每组8只。为期9 w,建模成功后,称量大鼠体重、脂肪重量,收集大鼠血液检测血脂四项、白细胞介素6(IL-6)、肿瘤坏死因子(TNFα)、胰岛素(insulin)和瘦素(leptin)的水平。取皮下脂肪,HE染色,观察脂肪细胞的形态,免疫印迹(Western blotting)方法测定脂肪分化蛋白BMP4、SMAD1、ZFP423、PPARγ、FABP4、LPL的表达。结果大鼠皮下脂肪量随着肥胖程度的增高而增加,与MC组相比,ML、MN、MS组的大鼠体重均显著升高(P<0.05);MN和MS组大鼠血清总胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白胆固醇(LDL-C)的水平显著升高(P<0.05);MN和MS组大鼠血清IL-6、TNFα、Insulin和Leptin浓度显著升高(P<0.05);与MC组相比,MN和MS组大鼠皮下脂肪细胞体积明显增大(P<0.05),细胞数量明显减少(P<0.05);MN和MS组大鼠皮下脂肪BMP4、SMAD1和ZNF423蛋白表达显著降低(P<0.05),ML、MN和MS组大鼠PPARγ和FABP4蛋白表达极显著降低(P<0.01)。结论不同程度的肥胖对大鼠皮下脂肪再分化过程均存在抑制作用。BMP4/PPARγ通路中不同阶段的调控蛋白对肥胖程度的敏感性不同,大鼠肥胖程度越高,皮下脂肪分化相关蛋白表达越低。Objective To observe the differentiation status of subcutaneous adipocytes in rats with different degrees of obesity induced by high-fat diet,and to explore the effect of different degrees of obesity on the BMP4/PPARγsignaling pathway of subcutaneous adipo cytes differentiation in rats.Methods SD rats were randomly divided into normal diet group(NC),high-fat diet group.The high-fat diet group were further divided into subgroups:overweight(ML),mild obesity(MN)and moderate obesity(MS).After the modeling was successful,body weight and fat weight were weighed,and blood samples were collected to detect blood lipids,interleukin-6(IL-6),tumor necrosis factor(TNFα),insulin and leptin.The subcutaneous fat was collected and stained with HE to observe the morphology of adipocytes.The protein expressions of BMP4,SMAD1,ZFP423,PPARγ,FABP4 and LPL in subcutaneous adipose tissue were determined by Western blotting.Results The amount of subcutaneous fat in rats increased with the degree of obesity.Compared to the NC group,the body weights of the rats in the ML,MN and MS groups were significantly increased(P<0.05).Serum levels of cholesterol(TC),triglyceride(TG)and low-density lipoprotein cholesterol(LDL-C)were significantly increased(P<0.05).Serum concentrations of IL-6,TNFα,insulin and leptin were significantly increased in the MN and MS groups(P<0.05).Compared to the NC group,the volume of subcutaneous adipocytes in the MN and MS groups was significantly increased(P<0.05),and the number of cells was significantly decreased.The protein expressions of BMP4,SMAD1 and ZNF423 in the subcutaneous fat of the MN and MS groups were significantly decreased(P<0.05),and PPARγand FABP4 protein expressions were significantly decreased in the ML,MN and MS groups(P<0.01).Conclusion Different degrees of obesity have inhibitory effects on the differentiation of subcutaneous adipocytes in rats.Different stages of regulatory proteins in the BMP4/PPARγpathway have different sensitivities to the degree of obesity.In the process of mature adipo
