摘  要:[目的]检测羊肉中是否含有貉肉成分。[方法]通过实时荧光定量PCR方法,以cytB为靶基因设计特异性检测引物,选取8个不同物种的肌肉组织样本为研究对象,根据其ΔCt值函数关系进行线性拟合建立标准曲线。[结果]该检测方法所用引物能将貉与其他物种区分,特异性较强,且最低检测限可达到3.2 pg/μL,回收率在97.71%~104.36%,组内变异系数≤0.28%,组间变异系数≤1.08%。[结论]该研究建立的羊肉中貉肉源成分实时荧光定量检测方法具有良好的特异性和敏感性,可用该方法检测实际羊肉样品中是否有貉肉源成分,为羊肉制品掺假的检测提供简单快捷准确的技术手段和执法依据。[Objective]To detect whether raccoon dog meat components are contained in mutton.[Method]We designed specific detection primers using cytB as a target gene by real-time PCR,selected muscle tissue samples from eight different species as study subjects,and established a standard curve by linear fitting according to theirΔCt value function relationship.[Result]The primers used in this detection method could distinguish raccoon from other species,with specificity,the lowest detection limit could reach 3.2 pg/μL,the recovery rate was 97.71%-104.36%,the intraclass coefficient of variation was≤0.28%,and the coefficient of variation was≤1.08%.[Conclusion]The real-time fluorescence quantitative detection method system for raccoon dog meat components in mutton established in this study has good specificity and sensitivity,and this method can be used to detect whether there are raccoon dog meat components in actual mutton samples,providing a simple,rapid and accurate technical means and law enforcement basis for the detection of adulteration of mutton products.
