摘　　要：[目的]探讨LncRNA PCAT1调控miR-128对肺癌H1299细胞放射敏感性的影响。[方法]用LncRNA PCAT1阻断剂处理或不处理肺癌H1299细胞,根据放射剂量的不同将细胞分为空白对照组(0Gy)、低剂量组(2Gy)、中剂量组(4Gy)和高剂量组(8Gy)。采用RT-qPCR检测各组细胞LncRNA PCAT1、miR-128表达水平,CCK-8法检测细胞存活率,transwell实验检测细胞侵袭力,划痕实验检测细胞迁移力,流式细胞术检测细胞凋亡率。[结果]空白对照组及各放射剂量组肺癌H1299细胞使用阻断剂后LncRNA PCAT1表达显著降低,miR-128显著升高(P<0.05)。与空白对照组比较,低、中、高剂量组肺癌H1299细胞存活率、迁移率和侵袭力显著降低,凋亡率显著升高(P<0.05)。使用阻断剂后各剂量组细胞存活率、迁移率和侵袭力均显著降低,凋亡率显著升高(P<0.05)。[结论]沉默lncRNA PCAT1可上调miR-128表达来增强肺癌H1299细胞的放射敏感性。[Objective]To investigate the effect of Silencing lncRNA PCAT1 on enhancing the radiosensitivity of lung cancer H1299 cells by regulating miR-128 expression.[Method]LncRNA PCAT1 blocker treated or not treated lung cancer H1299 cells,according to the different radiation doses,the cells were divided into blank control group(0 Gy),low dose group(2 Gy),middle dose group(4 Gy)and high dose group(8 Gy).RT-qPCR was used to detect the expression levels of LncRNA PCAT1 and miR-128 in each group of cells,CCK-8 method to detect cell viability,transwell assay to detect cell invasion,scratch assay to detect cell migration,flow cytometry to detect cell apoptosis rate.[Result]The expression of LncRNA PCAT1 in the blank control group and the lung cancer H1299 cells of each radiation dose group was significantly decreased,and miR-128 significantly increased(P<0.05).Compared with the blank control group,the survival rate of lung cancer H1299 cells in the low,medium and high dose groups,migration rate and invasiveness significantly decreased,apoptosis rate were significantly increased(P<0.05).After using the blocking agent,the cell survival rate of each dose group,migration rate and invasiveness were significantly decreased,and the apoptosis rate was significantly increased(P<0.05).[Conclusion]Silencing lncRNA PCAT1 can up-regulate miR-128 expression to enhance the radiosensitivity of lung cancer H1299 cells.
