摘 要:目的:研究NOD样受体C5(NLRC5)对肝癌细胞SMMC-7721凋亡及核转录因子-κB(NF-κB)表达水平的影响。方法:将NLRC5 miRNA转染至SMMC-7721细胞中使NLRC5低表达,并将SMMC-7721细胞分为对照组、Vector组和NLRC5组。比较3组细胞中NLRC5 mRNA的表达、凋亡率、增殖能力和NF-κB蛋白和相对表达量。结果:正常肝细胞中NLRC5 mRNA表达显著低于肝癌细胞。对照组和Vector组细胞中NLRC5 mRNA相比较无差异;与对照组相比较,NLRC5组细胞中NLRC5 mRNA的表达显著降低。与对照组比较,Vector组凋亡的肝癌细胞数量无差异;与对照组、Vector组相比,NLRC5组肝癌细胞的凋亡率显著增加。与对照组相比,Vector组细胞的增殖能力无变化;而NLRC5组细胞的增殖能力显著低于对照组和Vector组。与对照组和Vector组相比较,NLRC5组细胞中的NF-κB蛋白和NF-κB mRNA表达显著升高。结论:低表达NLRC5可促进肝癌细胞凋亡,其机制可能与负调控NF-κB有关,进而对肝癌的发生发展起到抑制作用。Objective:To study the effects of NOD-like receptor C5(NLRC5)on apoptosis and expression of nuclear factor-κB(NF-κB)in hepatoma SMMC-7721 cells.Methods:NLRC5 miRNA was transfected into SMMC-7721 cells for low expression of NLRC5,and SMMC-7721 cells were divided into three groups:a control group,a Vector group and a NLRC5 group.The expression of NLRC5 mRNA in three groups was compared.The apoptosis ability and proliferation ability of the three groups were compared.NF-κB relative expression were compared among the three groups.Results:NLRC5 mRNA expression in normal hepatocytes was significantly lower than that in hepatocelluar carcinma(HCC).There was no difference in NLRC5 mRNA between the control group and Vector group.Compared with the control group,the expression of NLRC5 mRNA in NLRC5 group was significantly decreased.Compared with the control group,there was no difference in the number of apoptotic HCC cells in the Vector group.Compared with control group and Vector group,apoptosis number and apoptosis rate of HCC in NLRC5 group were significantly increased.Compared with the control group,the proliferation ability of cells in the Vector group did not change.The proliferation ability of NLRC5 group was significantly lower than that of control group and Vector group.NF-κB protein and NF-κB mRNA expression were significantly increased in NLRC5 group compared with control group and Vector group.Conclusion:Low expression of NLRC5 can promote HCC apoptosis,which may be related to the negative regulation of NF-κB,and thus inhibit the occurrence and development of hepatocellular carcinoma.
