摘 要:目的探究小鼠海马齿状回和前扣带回区域静息状态的神经活动与酒精依赖的相关性。方法21只小鼠进行了为期1个月的自由酒精摄取造模,对所有小鼠进行酒精偏好性检测、灌注取脑,进行早期报告基因c-fos的免疫组化染色。结果21只小鼠均形成了不同程度的酒精依赖(酒精偏好大于50%):其中5只形成较低程度的酒精依赖(酒精偏好小于60%),5只形成较高程度的酒精依赖(酒精偏好大于70%);小鼠海马齿状回区域的c-fos表达水平和酒精偏好呈负相关性(r^(2)=0.2151,P=0.0342),且低酒精偏好小鼠海马齿状回区域的c-fos表达高于高酒精偏好小鼠(T8=2.537,P=0.0349);前扣带回区域的c-fos表达和酒精偏好无相关性(r^(2)=2.86*10-5,P=0.9818)。结论小鼠海马齿状回可能在酒精依赖中发挥作用,针对海马齿状回的治疗可能对于重度酒精依赖患者有帮助。Objective To explore whether the neural activity of hippocampal dentate gyrus(DG)and anterior cingulate cortex(ACC)play a role in alcohol dependence in mice.Methods Twenty-one mice were freely exposed to alcohol intake for one month and were tested for alcohol preference.Immunohistochemical staining of immediate early gene c-fos was performed on the brain of the mice after perfusion.Results All the 21 mice developed different degree of alcohol dependence(alcohol preference>50%),among which 5 mice developed low degree of alcohol dependence(alcohol preference<60%),5 mice developed high degree of alcohol dependence(alcohol preference>70%).The expression level of c-fos in the DG of the hippocampus was negatively correlated with alcohol preference(r^(2)=0.2151,P=0.0342),and significantly difference c-fos level between low and high alcohol preference mice(T=2.537,P=0.0349)in the DG.There was no correlation between c-fos expression in ACC and alcohol preference(r^(2)=2.86*10,P=0.9818).Conclusion The DG may play a role in alcohol dependence in mice,and the regulation of the DG may be helpful for patients with severe alcohol dependence.
