摘 要:目的:比较大脑中动脉闭塞(middle cerebralartery occlusion,MCAO)大鼠不同缺血再灌注时间术后部分功能状况,为MCAO模型缺血时间选择提供参考。方法:36只Sprague-Dawley(SD)大鼠采用改良线栓法制备MCAO模型,随机分为缺血再灌注1 h组、1.5 h组、2 h组。通过体质量、Longa评分、平衡木功能评分、脑梗死体积等指标,分析3组大鼠术后状况。结果:1.5 h组、2 h组大鼠体质量第7天较第1天体质量明显下降(P<0.05):1.5 h组、2 h组大鼠Longa评分第7天较第1天升高,且较同时段lh组相比得分高(P<0.05);1 h组大鼠平衡木评分第7天较第1天降低(P<0.05),1.5h组、2h组平衡木评分在第3天、第7天比同时段1 h组较高(P<0.05);TTC染色可见1.5 h组、2h组大鼠脑梗死体积大于1 h组(P<0.05)。结论:1 h组大鼠脑损伤程度低,术后神经功能、运动功能恢复快:1.5 h组与2 h组各项评分结果相似,其功能状况无明显差别,但与1 h组比脑损伤严重。1.5 h和2 h更适合作为线栓法制备MCAO模型的再灌注时间点。Objective:To compare the partial functional status of rats with middle cerebral artery occlusion(MCAO) after different ischemiareperfusion time,so as to provide reference for the selection of ischemia time of MCAO model.Methods:36 Sprague Dawley(SD) rats were randomly divided into ischemia-reperfusion 1h group,reperfusion 1.5 h group and reperfusion 2 h group.The postoperative conditions of the three groups were analyzed the success rate of rat modeling,body mass,longa score,balance beam function score,cerebral infarct volume and so on.Results:The body mass of rats in 1.5 h group and 2 h group decreased significantly on the 7th day compared with the 1st day(P<0.05);The longa score of rats in 1.5 h group and 2 h group on the 7th day was higher than that in the 1st day,and higher than that in 1 h group at the same time(P<0.05);On the 7th day,the score of balance beam in the 1 h group was lower than that in the 1st day(P<0.05),and on the 3rd and 7th day,the score of balance beam in the 1.5 h group and the 2 h group was higher than that in the 1 h group at the same time(P<0.05);TTC staining showed that the volume of cerebral infarction in 1.5 h group and 2 h group was larger than that in 1h group(P<0.05).Conclusion:In the 1 h group,the degree of brain injury was mild,and the neurological and motor functions recovered quickly after operation;the scores of 1.5 h group and 2 h group were similar,and there was no significant difference in their functional status,but the brain injury was more serious than 1 h group.1.5 h and 2 h can be used as the reperfusion time points of MCAO model prepared by thread bolt method.
