摘  要:目的多巴胺含量的测定对预测生物体的行为表现具有重要意义。方法采用酶催化荧光光谱分析法,利用多巴胺对反应产物荧光强度的猝灭,实现了对尿液中多巴胺的检测。结果通过探讨检测体系的荧光特性、稳定性、反应物的浓度、试剂的加入顺序等因素对测定结果的影响,最终选择在最大激发305 nm,最大发射401 nm处进行测定,荧光猝灭强度与多巴胺浓度在10~50μg·mL^(-1)浓度范围内存在良好的线性关系,线性回归方程为y=0.536x+68.338,R^(2)=0.9876,回收率为96.17%~99.01%。结论该方法具有操作简便、耗时短、可靠性高等优点,可应用于尿液中多巴胺的测定。OBJECTIVE Dopamine is a neurotransmitter secreted by the brain and its levels are important in predicting the behavioural performance of an organism.By exploring the detection of dopamine content in urine,it is of great significance and value for the diagnosis and detection of related diseases.METHODS In this experiment,an enzyme-catalyzed fluorescence spectroscopy method was used to detect dopamine in urine by using the burst of dopamine on the fluorescence intensity of the reaction products.The effects of the fluorescence characteristics of the system,its stability,the concentration of the reactants,the order of addition of reagents on the determination results were explored.RESULTS The final determination was chose at a maximum excitation of 305 nm and a maximum emission of 401 nm.A good linear relationship existed between the fluorescence burst intensity and the concentration of dopamine in the concentration range of 10-50μg·mL^(-1).The linear regression equation was y=0.536x+68.338,R^(2)=0.9876 and the recoveries were 96.17%~99.01%.CONCLUSION The method has the advantages of simple operation,short time consumption and high reliability and can be applied to the determination of dopamine in urine.
