摘 要:为寻找一种方便、稳定、快速的途径来评估抗氧化剂的抗氧化能力,采用一种简单、经济的方法制备负载普鲁士蓝(PB)的羟基氧化钴纳米片(PB-CoOOH NFs)。以对苯二甲酸(TA)为荧光探针研究了PB-CoOOH NFs酶促反应的机制,基于PB-CoOOH NFs/TMB/H_(2)O_(2)体系对10种常见的抗氧化剂进行了抗氧化活性测定(TMB为3,3′,5,5′-四甲基联苯胺)。结果表明,CoOOH纳米片的过氧化物模拟酶活性随着普鲁士蓝的引入而增强。相比天然酶(HRP),PB-CoOOH NFs具有更小的Km值,对底物的亲和力更高。在一定浓度范围内,10种抗氧化剂测得的吸光度差与浓度呈良好的线性关系,相关系数(R2)均大于0.994,检出限为0.082~0.315μmol/L,与其他检测抗氧化活性的方法相比灵敏度更高。In order to seek for a convenient,stable and rapid way to evaluate the antioxidative capacity of antioxidant,Prussian blue(PB)loaded cobalt oxyhydroxide nanoflakes(PB-CoOOH NFs)are prepared through a simple and economical method.The mechanism of enzymatic reaction of PB-CoOOH NFs is investigated with terephthalic acid(TA)as a fluorescent probe.The antioxidative activity of 10 kinds of common antioxidants are determined based on PB-CoOOH NFs/TMB/H_(2)O_(2)system(TMB is 3,3’,5,5’-tetramethylbenzidine).The results show that the peroxidase-mimicking activity of CoOOH can be enhanced with the addition of PB.Compared with natural enzymes(HRP),PB-CoOOH NFs has smaller Kmvalue and higher affinity for substrates.The measured absorbance differences of the 10 kinds of antioxidants show a good linearity with their concentrations in a certain concentration range,and the correlation coefficients(R^(2))are all greater than 0.994.The detection limits range from 0.082 to 0.315μmol·L^(-1),showing a more sensitivity than other methods for the detection of antioxidative activity.
