摘  要:目的探究5-氨基乙酰丙酸(5-ALA)光动力疗法(PDT)对裸鼠人上皮性卵巢癌移植瘤生长的抑制作用。方法建立裸鼠人上皮性卵巢癌皮下移植瘤模型,分为模型组、5-ALA瘤注组、5-ALA静注组、PDT组、5-ALA瘤注-PDT组、5-ALA静注-PDT组,10只/组。模型组、PDT组给予瘤内注射无菌生理盐水溶液,5-ALA瘤注组、5-ALA瘤注-PDT组给予瘤内注射5-ALA(40 mg/kg)生理盐水溶液,5-ALA静注组、5-ALA静注-PDT组给予尾静脉注射5-ALA(40 mg/kg)生理盐水溶液。1 h避光后,PDT组、5-ALA瘤注-PDT组、5-ALA静注-PDT组用630 nm激光治疗仪局部照射瘤体治疗20 min,每隔2天各组治疗1次,共5次。治疗完成后,安乐处死各组小鼠,肿瘤组织HE染色,测量瘤体体积,采用TUNEL染色法检测肿瘤细胞凋亡率,采用实时定量PCR(qRT-PCR)检测组织cleaved Caspase-3、Bcl-2与Bax的mRNA表达水平,采用Western blot检测组织cleaved Caspase-3、Bcl-2与Bax蛋白表达水平,计算各组的脏器系数。结果相比于模型组、5-ALA瘤注组、5-ALA静注组,PDT组、5-ALA瘤注-PDT组和5-ALA静注-PDT组干预14 d后的瘤体体积均减小,且5-ALA瘤注-PDT组的瘤体体积最小(均P<0.05)。治疗7、14 d后的抑瘤率在PDT组、5-ALA瘤注-PDT组和5-ALA静注-PDT组高于模型组、5-ALA瘤注组、5-ALA静注组(均P<0.05),并且5-ALA瘤注-PDT组的抑瘤率最高(均P<0.05)。组织cleaved Caspase-3、Bax的mRNA与蛋白表达在PDT组、5-ALA瘤注-PDT组和5-ALA静注-PDT组高于模型组、5-ALA瘤注组、5-ALA静注组(均P<0.05),5-ALA瘤注-PDT组的cleaved Caspase-3、Bax的mRNA与蛋白表达量最高(均P<0.05)。组织Bcl-2的mRNA与蛋白表达在PDT组、5-ALA瘤注-PDT组和5-ALA静注-PDT组低于模型组、5-ALA瘤注组、5-ALA静注组(均P<0.05),5-ALA瘤注-PDT组组织Bcl-2的mRNA与蛋白表达水平最低(均P<0.05)。心脏、肝脏、脾脏和肺的脏器系数在PDT组、5-ALA瘤注-PDT组高于模型组、5-ALA瘤注组、5-ALA静注组(均P<0.05),5-ALA静注-PDT组�Objective To explore the inhibitory effect of 5-ALA photodynamic therapy(PDT)on the growth of human epithelial ovarian cancer grafts in nude mice.Methods Human epithelial ovarian cancer subcutaneous transplantation tumor models were established and divided into the model group,5-ALA tumor injection group(5-ALA TI group),5-ALA intravenous injection group(5-ALA IV group),PDT group,5-ALA tumor injection-PDT group(5-ALA TI-PDT group)and 5-ALA intravenous injection-PDT group(5-ALA IV-PDT group).There were 10 rats in each group.The Model group and PDT group were given intratumoral injection of sterile saline solution,the 5-ALA TI group and 5-ALA TI-PDT group were given intratumoral injection of 5-ALA solution(40 mg/kg),5-ALA IV group and 5-ALA IV-PDT group were given 5-ALA solution via tail vein injection(40 mg/kg).After being protected from light for 1 h,the PDT group,5-ALA TI-PDT group and 5-ALA IV-PDT group were treated with 630 nm laser therapy device for local irradiation of the tumor for 20 min,once per 2 d,5 times in total.After the treatment,the mice in each group were euthanized.The tumor tissues were collected,and HE staining was used to measure the tumor volume.The TUNEL staining was used to detect the apoptosis rate of tumor cells.The real-time quantitative PCR(qRT-PCR)was used to detect the cleaved Caspase-3,Bcl-2 and Bax mRNA expression in tumor tissue.The Western blot was used to detect the expression of cleaved Caspase-3,Bcl-2 and Bax protein in tumor tissue.The organ coefficient of each group was calculated.Results Compared with the model group,the tumor volumes in the 5-ALA TI group,5-ALA IV group,PDT group,5-ALA TI-PDT group and 5-ALA IV-PDT group after 14 d of treatment were lower(all P<0.05),and the 5-ALA TI-PDT group had the lowest tumor volume(all P<0.05).After 7 d and 14 d of treatment,the tumor inhibition rates in the PDT group,5-ALA TI-PDT group and 5-ALA IV-PDT group were higher than those in the model group,5-ALA TI group and 5-ALA IV group(all P<0.05),and the 5-ALA TI-PDT group had the high
