摘  要:近年来,随着医疗技术的不断进步,接受颅脑照射治疗的肿瘤患者的长期生存率大幅提升。因此,颅脑照射的远期并发症,如智力下降、记忆力减退、甚至是痴呆等问题愈来愈受到人们的重视。糖原合酶激酶-3(glycogen synthase kinase-3,GSK-3)是一种高度保守的丝氨酸/苏氨酸蛋白激酶,广泛存在于真核生物的细胞质中,目前已发现有GSK-3α和GSK-3β两种亚型。GSK-3β在脑组织尤其是海马区域中高表达,广泛参与各种神经退行性疾病、神经精神类疾病和癌症等多种重大疾病的发病过程。研究显示GSK-3β参与细胞炎症反应、糖原代谢、神经系统功能等的调控,激活GSK-3β的活性会影响整个生命周期中的细胞增殖、衰老、凋亡及突触的可塑性,进而导致神经系统认知功能的障碍。目前,GSK-3β与颅脑照射后认知障碍的相关性研究较少,该文结合国内外近年来的研究成果对GSK-3β在颅脑照射后认知功能障碍中的作用展开综述。The long-term survival rate of tumor patients treated with cranial irradiation has increased dramatically with the continuous advancement of medical technology in recent years.As a result,the long-term complications of cranial irradiation,such as mental decline,memory impairment,and even dementia,are gaining more and more attention.Glycogen synthase kinase-3(GSK-3)is a highly conserved serine/threonine protein kinase that is widely found in the cytoplasm of eukaryotes.And two isoforms,GSK-3αand GSK-3β,have been identified.GSK-3βis highly expressed in brain tissue,especially in the hippocampus,and is widely involved in the pathogenesis of various neurodegenerative diseases,neuropsychiatric disorders and cancer.Studies have shown that GSK-3βis involved in the regulation of cellular inflammatory response,glycogen metabolism,neurological function,etc.The activation of GSK-3βaffects cell proliferation,aging,apoptosis,and synaptic plasticity throughout the life cycle,leading to neurological cognitive dysfunction.At present,there are few studies on the correlation between GSK-3βand cognitive impairment after cranial irradiation.This paper reviews the role of GSK-3βin cognitive dysfunction after cranial irradiation with reference to domestic and foreign studies.
