摘 要:目的:利用核磁共振氢谱(^(1)H-NMR)代谢组学方法对慢性温和不可预知应激(CUMS)抑郁大鼠粪便进行研究,从粪便代谢组学角度探讨百合鸡子汤干预CUMS大鼠代谢轮廓的作用机制。方法:CUMS法复制大鼠抑郁模型,对造模好的大鼠粪便进行前处理,采用^(1)H-NMR技术检测粪便中的代谢物质的图谱,对图谱进行解析得出相应数据,再对数据进行多元统计分析,研究代谢物轮廓的变化。结果:多元数据统计分析可知,空白组和抑郁模型组散点图差异非常明显,表明造模成功。从大鼠粪便中一共分析得到14种代谢物质,经过差异筛选得到谷氨酸、乳酸两种关键代谢物质。这两种关键差异性代谢物在正常大鼠和CUMS抑郁大鼠粪便中的含量存在明显差异,而中药组的谷氨酸含量比模型组显著增加,乳酸含量比模型组降低。结论:百合鸡子汤可能通过调节谷氨酸和乳酸代谢对抑郁症产生改善作用。Objective:^(1)H-NMR metabonomics was used to study the Chronic mild and unpredictable stress(CUMS) rat model, and the effect of Baihe Jizi Decoction(BHT) on the anti-depression of CUMS rats was investigated from the perspective of metabonomics.Methods:The pre-molded rat feces were processed, and the metabolite atlas in feces was detected by ^(1)H-NMR technology.The atlas was analyzed to obtain the corresponding data, and then multivariate statistical analysis was conducted on the data to study the change of metabolite.Results:According to the statistical analysis of the multivariate data of rats, the difference of scatter plots between the control group and the CUMS group was very obvious, indicating that the modeling was successful.A total of 14 metabolites were analyzed from the feces of rats, among which two key metabolites, glutamic acid and lactic acid, were analyzed.To some extent, it can be explained that there was a significant content difference in fecal between contral rats and CUMS rats.The content of Glutamate in the CUMS+BHT group was higher than that in the CUMS group, while the content of lactic acid in the CUMS group was higher than that in the CUMS+BHT group, which provides certain reference for the future study of rat depression.Conclusion:Baihe Jizi Decoction may have an ameliorated effect on depression by regulating glutamate and lactate metabolism.
