摘 要:以卡氏芽孢杆菌(Bacillus cabrialesii)ST-1为研究对象,芽孢数为响应值,采用单因素试验、Plackett-Burman试验、最陡爬坡试验及响应面试验对其发酵培养基进行优化。结果表明,影响卡氏芽孢杆菌ST-1芽孢数的主要因素为蔗糖、麸皮和蛋白胨、磷酸二氢钾,卡氏芽孢杆菌ST-1产芽孢的最适宜培养基配方为:蔗糖13.1 g/L,麸皮+蛋白胨(1∶2)17.0 g/L,磷酸二氢钾2.0 g/L。在此最优条件下,卡氏芽孢杆菌ST-1发酵液中芽孢数达到5.96×10^(9) CFU/mL,是优化前的32.21倍。Using Bacillus cabrialesii ST-1 as the research object,and spore number as the response value,the fermentation medium was optimized by single factor tests,Plackett-Burman tests,steepest climb tests,and response surface tests.The results showed that the main factors affecting the spore number of B.cabrialesii ST-1 were sucrose,bran and peptone,and potassium dihydrogen phosphate.The optimal medium formula for spore production by B.cabrialesii ST-1 was as follows:sucrose 13.1 g/L,bran+peptone(1∶2)17.0 g/L,potassium dihydrogen phosphate 2.0 g/L.Under the optimal conditions,the spore number in the fermentation broth of B.cartesii ST-1 reached 5.96×10^(9) CFU/ml,which was 32.21 times that of before optimization.
