海参筋中海参皂苷提取分离及结构分析

摘  要:海参皂苷是由苷元和糖基侧链组成的结构复杂的糖基类化合物.本研究以海参筋为原料进行海参皂苷的提取,以D101大孔吸附树脂和半制备液相色谱仪进行分离纯化,得到3种物质SCTS-Ⅳ-W、SCTS-Ⅳ-20和SCTS-Ⅳ-40.采用傅里叶变换红外光谱(FTIR)、气相色谱法(GC)以及安装了电喷雾电离源和飞行时间离子阱的串联质谱仪(ESI-IT/TOF-MS/MS)分析其结构,结果表明:3种物质含有皂苷类物质的官能团以及奎诺糖、木糖、3-O-甲基葡萄糖和葡萄糖4种糖基,并且发现了其可能的结构.Sea cucumber saponins are glycosyl compounds with complex structures composed of aglycones and glycosyl side chains.In this study,sea cucumber saponins were extracted from sea cucumber tendon as raw material,purified and separated by D101 macroporous adsorption resin and semi-preparative liquid chromatography,and three substances SCTS-Ⅳ-W,SCTS-Ⅳ-20 and SCTS-Ⅳ-40 were obtained.The structures were analyzed by Fourier transform infrared spectroscopy(FTIR),gas chromatography(GC)and tandem mass spectrometer with electric spray ionization and time-of-flight ion trap(ESI-IT/TOF-MS/MS).The results showed that the three substances contained the functional groups of saponins,quinose,xylose,3-O-methyl glucose and glucose 4 glycosyl groups,and their possible structures were also found.
