摘  要:目的探讨黄精酒制前后多糖的单糖组成差异及抗氧化活性的变化。方法采用L9(34)正交表进行正交试验,确定黄精多糖的最佳水解条件,并对比高效液相色谱-蒸发光散射检测法(High Performance Liquid Chromatography-Evaporative Light Scattering Detection,HPLC-ELSD)、高效液相色谱-示差折光检测法(High Performance Liquid Chromatography-Refractive Index Detector,HPLC-RID)和1-苯基-3-甲基-5-吡唑啉酮柱前衍生化高效液相色谱仪-二极管阵列检测法(3-Methyl-1-phenyl-2-pyrazolin-5-one-High Performance Liquid Chromatography-Diode Array Detector,PMP-HPLC-DAD)3种分析方法的检测效果,确定测定黄精多糖单糖组成的最佳检测方法,考察黄精酒制前后多糖的单糖组成变化,并运用主成分分析法(Principal Components Analysis,PCA)和正交偏最小二乘辨别分析法(Orthogonal Partial Least Squares-Discriminant Analysis,OPLS-DA)进行数据统计,寻找差异性成分。最后以1,1-二苯基-2-三硝基苯肼(1,1-Diphenyl-2-Picrylhydrazyl,DPPH)自由基清除率大小为指标考察黄精酒制前后多糖的体外抗氧化活性。结果对比3种分析方法,HPLC-ELSD的检测效果最好,可检测到阿拉伯糖、果糖、甘露糖和葡萄糖4种单糖。PCA和OPLS-DA结果均显示黄精酒制前后单糖含量存在差异,可明显区分开,并筛选出3个差异性成分。此外,黄精酒制后对DPPH自由基的清除能力增强,半数抑制浓度(IC_(50))为0.57 mg/mL。结论采用HPLC-ELSD法的检测效果最好,该方法简单方便,重复性好。酒制对黄精多糖的单糖组成影响较大,黄精酒制后多糖发生水解及结构发生改变,其抗氧化活性增强。该研究可为黄精多糖的开发利用奠定一定的基础。Objective To investigate the differences of monosaccharide composition and changes of antioxidant activity from Polygonatum Sibiricum(PS) and Processed Polygonatum Sibiricum(PPS) polysaccharides.Methods The orthogonal test was performed using L9(34) orthogonal table to determine the optimal hydrolysis conditions for PS and PPS polysaccharides.The testing results of three analytical methods including HPLC-ELSD,HPLC-RID and PMP-HPLC-DAD were compared to determine the best method for detecting the monosaccharide composition of PS and PPS polysaccharides,exploring the changes of the monosaccharide composition of PS and PPS polysaccharides.Meanwhile,Principal Component Analysis(PCA) and Orthogonal Partial Least Squares-Discriminant Analysis(OPLS-DA) were used to conduct statistical analysis,looking for the differential components.The antioxidant activities of PS and PPS polysaccharides were studied by 1,1-Diphenyl-2-Picrylhydrazyl(DPPH) radical scavenging ability as an indicator.Results Comparing the three analytical methods,the HPLC-ELSD method was best,which can detect four monosaccharides including arabinose,fructose,mannose and glucose.PCA and OPLS-DA results showed the monosaccharide content of PS and PPS polysaccharides existed some differences,which could be distinguished obviously,and three differential components were screened out.In addition,the DPPH radical scavenging ability of PPS polysaccharides significantly enhanced compared to PS polysaccharides,and the half maximal inhibitory concentration(IC_(50)) was 0.57 mg/mL.Conclusion The HPLC-ELSD method was the best for the detection of monosaccharide composition,which was simple,convenient and reproducible.Processing with wine had a strong effect on the monosaccharide composition of PS polysaccharides,and we observed that Polysaccharides happened hydrolysis and structural changes after processing,and their antioxidant activities enhanced.This study can establish a certain foundation for the development and utilization of the Polygonatum Sibiricum polysacchar
