摘  要:目的 探讨miRNA-455-3p在乳腺癌组织中的表达及与紫杉醇(PTX)耐药的关系。方法 选取80例乳腺癌患者,采用实时定量逆转录聚合酶链反应(qRT-PCR)检测乳腺癌组织和癌旁组织中miRNA-455-3p的相对表达量及PTX耐药和PTX敏感乳腺癌患者乳腺癌组织中miRNA-455-3p、自噬相关蛋白(ATG12)mRNA的相对表达量。将miRNA-NC、miRNA-455-3p mimics分别转染至乳腺癌耐药MCF-7/PTX细胞,分别作为miRNA-NC组、miRNA-455-3p组,采用蛋白质印迹法检测细胞凋亡情况以及P糖蛋白(P-gp)和谷胱甘肽S-转移酶-π(GST-π)蛋白的相对表达量。相关性分析采用Spearman相关分析。结果 乳腺癌组织中miRNA-455-3p的相对表达量明显高于癌旁组织,差异有统计学意义(P﹤0.01)。PTX敏感乳腺癌患者乳腺癌组织中miRNA-455-3p的相对表达量明显高于PTX耐药患者,ATG12 m RNA相对表达量明显低于PTX耐药患者,差异均有统计学意义(P﹤0.01)。miRNA-455-3p组细胞凋亡率明显高于miRNA-NC组,P-gp、GST-π蛋白相对表达量均明显低于miRNA-NC组,差异均有统计学意义(P﹤0.01)。Spearman相关分析结果显示,乳腺癌组织中miRNA-455-3p的表达与ATG12的表达呈负相关(r=-0.697,P﹤0.01)。结论 乳腺癌组织miRNA-455-3p的表达与ATG12的表达呈负相关,通过靶向抑制ATG12的表达降低耐药乳腺癌细胞的自噬活性,促进细胞凋亡,有助于改善乳腺癌细胞对PTX药物的敏感性。Objective To investigate the expression of miRNA-455-3p in breast cancer tissue and its relationship with paclitaxel(PTX) resistance. Method A total of 80 breast cancer patients were selected. Quantitative reverse transcription-polymerase chain reaction(qRT-PCR) was used to detect the relative expression of miRNA-455-3p in breast cancer tissue and adjacent tissue, the relative expression of miRNA-455-3p and autophagy related 12(ATG12) mRNA in PTX-sensitive and PTX-resistant breast cancer tissues. The miRNA-NC and miRNA-455-3p mimics were transfected into breast cancer drug-resistant MCF-7/PTX cells, respectively, as the miRNA-NC group and miRNA-455-3p group. Apoptosis and the relative expression of P-glycoprotein(P-gp) and glutathione S-transferase-π(GST-π) protein were detected by Western blot. Correlation analyses were based on the Spearman correlation analysis. Result The relative expression of miRNA-455-3p in breast cancer tissue was significantly higher than that in adjacent tissue(P<0.01). The relative expression of miRNA-455-3p in PTX-sensitive breast cancer tissue was significantly higher than that in PTX-resistant breast cancer tissue, and the relative expression of ATG12 mRNA was significantly lower than that in PTX-resistant brest cancer tissue, and the differences were statistically significant(P<0.01). The apoptosis rate of the miRNA-455-3p group was significantly higher than that of the miRNA-NC group, and the relative expression of P-gp and GST-π protein were significantly lower than those of the miRNA-NC group, and the differences were statistically significant(P<0.01). Spearman correlation analysis showed that the expression of miRNA-455-3p in breast cancer tissue was negatively correlated with the expression of ATG12(r=-0.697, P<0.01). Conclusion The expression of miRNA-455-3p in breast cancer tissue is negatively correlated with the expression of ATG12.
