摘  要:目的 通过微小免疫磁珠法制备神经干细胞(NSC),观察其治疗脊髓损伤(SCI)大鼠的效果。方法 将微小免疫磁珠分离纯化出神经巢蛋白(nestin)阳性的NSC移植到SCI的大鼠模型中。将30只Wister雌性大鼠随机分为实验组、空白对照组、培养液组,每组10只。实验组接受NSC移植,培养液组接受培养液注射,空白对照组接受生理盐水注射。采用运动能力评定量表(BBB)系统评估SCI后、NSC移植后大鼠运动能力。结果 术后10周取材,肉眼可见对照组损伤节段脊髓及其周围组织破坏程度高于实验组,空白对照组和培养液组肉眼下观察无明显区别。SCI模型构建14 d后3组大鼠的BBB评分符合SCI。实验组28~70 d的BBB评分高于空白对照组、培养液组(P<0.05);空白对照组和培养液组BBB评分全程组间比较,差异无统计学意义(P>0.05)。实验组大鼠28~70 d在斜板上停留5 s的平均最大角度均大于空白对照组和培养液组(P<0.05);空白对照组和培养液组大鼠28~70 d在斜板上停留5 s的平均最大角度比较,差异无统计学意义(P>0.05)。结论 采用微小免疫磁珠法制成的NSC移植到SCI大鼠内,可促进大鼠运动能力恢复。Objective Neural stem cells(NSCs) were prepared by micro-immune magnetic beads method to observe the therapeutic effect of NSCs on rats with spinal cord injury(SCI).Methods The nestin-positive NSC isolated and purified by micro-immune magnetic beads was transplanted into the rats model of SCI. Thirty Wister female rats were randomly divided into test group, blank control group and culture medium group, with 10 cases in each group. The test group received NSC transplantation, the culture solution group received culture solution injection, and the blank control group received physiological saline injection. The motor ability of rats after SCI and NSC transplantation was evaluated with the Basso, Beattie, Bresnahan(BBB).Results At 10 weeks after operation, the damage degree of the injured spinal cord and its surrounding tissues in the control group was higher than that in the test group, and there was no difference between the blank control group and the culture solution group. The BBB scores of the three groups of rats were consistent with SCI 14 days after the establishment of the SCI model. The BBB score of the test group from 28 to 70 days was higher than that of the blank control group and the culture medium group(P<0.05). There was no difference in BBB scores between the blank control group and the culture medium group(P>0.05). The average maximum angle of rats in the test group staying on the inclined plate for 5 s from 28 to 70 days was greater than that in the blank control group and the culture medium group(P<0.05). There was no statistically difference between the average maximum angle of rats in the blank control group and the culture solution group when they stayed on the inclined plate for 5 s from 28 to 70 days(P>0.05).Conclusion NSC prepared by micro-immune magnetic beads can be transplanted into SCI rats, which can promote the recovery of motor ability of rats.
