摘 要:目的分析乳腺癌癌组织增殖细胞核抗原(PCNA)、小鼠双微体扩增基因(MDM2)表达与临床病理特征及人类乳头状瘤病毒(HPV)16/18型感染的相关性。方法选取2018年3月-2021年3月沈阳市第四人民医院收治的乳腺癌患者117例,使用原位杂交法与免疫组化EnVision二步法检测乳腺癌组织与癌旁正常组织中HPV16/18、PCNA、MDM2及p53的表达,并分析PCNA、MDM2、p53表达与乳腺癌临床病理特征及HPV16/18感染的相关性。结果TNM分期高、肿瘤直径大及存在淋巴结转移的乳腺癌患者HPV16/18、PCNA、MDM2阳性率均高于TNM分期低、肿瘤直径小及不存在淋巴结转移的患者(P<0.05);Spearman相关性分析显示,PCNA、MDM2及p53表达与HPV16/18阳性率均呈正相关(P<0.05)。结论PCNA、MDM2在乳腺癌患者癌组织中呈高表达,且PCNA、MDM2、p53表达均与乳腺癌的发生发展及HPV16/18感染存在关联性,提示PCNA、MDM2可作为乳腺癌病情预测指标。OBJECTIVE To explore the association between expressions of proliferating cell nuclear antigen(PCNA)and murine double minute 2(MDM2)in breast cancer tissues and clinicopathological characteristics,human papilloma virus(HPV)type 16/18 infection.METHODS Totally 117 breast cancer patients who were treated in the Fourth People′s Hospital of Shenyang from Mar 2018 to Mar 2021 were enrolled in the study.The expressions of HPV16/18,PCNA,MDM2 and p53 in breast cancer tissues and adjacent normal tissues were detected by in situ hybridization and immunohistochemistry EnVision two-step method,and the association between the expressions of PCNA,MDM2,p53 and the clinicopathological characteristics,HPV16/18 infection was observed.RESULTS The positive rates of HPV16/18,PCNA and MDM2 of the breast cancer patients with high TNM stage,large diameter of tumor and lymphatic metastasis were higher than those of the breast cancer patients with low TNM stage,small diameter of tumor and without lymphatic metastasis(P<0.05).Spearman correlation analysis showed that the expressions of PCNA,MDM2 and p53 were positively correlated with the positive rate of HPV16/18(P<0.05).CONCLUSION The breast cancer patients show high expressions of PCNA and MDM2 in breast cancer tissues,and the expressions of PCNA,MDM2 and p53 are associated with the genesis and progression of breast cancer as well as the HPV16/18 infection,indicating that PCNA and MDM2 can be used as predictive indexes for the state of breast cancer.
