摘 要:为优化桑黄子实体中多糖含量的测定方法,本研究采用单因素试验,分别考察了桑黄子实体的粉碎粒度、提取时间、提取次数、乙醇体积等提取条件对桑黄子实体多糖提取率的影响,在确定最优提取条件后,进一步考察该提取条件下用硫酸蔥酮法测定桑黄多糖含量的精密度、重复性、回收率、中间精密度参数,最后利用该提取测定方法测定人工栽培桑黄产品的多糖含量。结果显示,桑黄子实体多糖的最优提取测定方法为桑黄子实体粉碎过50目筛,每2 g样品沸水浴加热回流4 h,水提次数2次,合并滤液后置水浴上蒸干,残渣用5 mL水溶解,加75 mL乙醇,4℃沉淀12 h。该条件下用硫酸蒽酮法测定同批次桑黄样品的精密度、重复性、回收率、中间精密度,结果均良好,相对标准偏差均小于3%。用该方法测定浙江省内10份不同来源或不同批次桑黄样品多糖含量,质量分数范围为1.92%~2.34%。研究结果可为浙江产区桑黄的多糖含量测定提供方法学依据和参考值。In order to optimize the method for the determination of polysaccharides content in Sanghuang fruiting body,a single factor test was conducted to investigate the effects of factors such as grinding size,extraction time,extraction times and ethanol volume on the extraction rate of polysaccharides from Sanghuang fruiting body.Under the selected optimal extraction conditions,the precision,repeatability,recovery rate and intermediate precision of polysaccharides determination method by using anthrone-sulfuric acid assay were further investigated.Finally,the method was used to determine the content of polysaccharides in artificially cultivated Sanghuang samples.The results showed that the optimal extraction and determination method of polysaccharide was as follows:dry grinded Sanghuang fruiting bodies powder was sifted through 50 mesh sieve,heated in boiling water bath for 4 h per 2 g sample and extracted twice.The combined filtrate was evaporated and dissolved in 5 mL water,and 75 mL ethanol was added for precipitation at 4℃for 12 h.Under these extraction conditions,the RSD of the precision,repeatability,recovery and intermediate precision of the same batch of Sanghuang sample were all less than 3%.The polysaccharide content of 10 batches of Sanghuang samples from different sources in Zhejiang Province determined by this method was 1.92%-2.34%.Our results provide methodological basis and reference values for the determination of polysaccharide content of Sanghuang in Zhejiang Province.
