摘 要:目的:探察和证实以场景恐惧记忆为模型建立的创伤后应激障碍(PTSD)状态下海马脑区的形态学、神经递质的改变,并探查其改变的相关调控机制。方法:30只C57雄性小鼠被随机分为2组,借助单纯场景恐惧记忆模型构建创伤后应激障碍、高尔基组织病理学染色、试剂盒方法检测海马区的形态学、神经递质和关键酶酶活的改变。结果:相较于正常对照组,形成恐惧记忆模型的小鼠海马区树突棘的密度增加,海马脑区谷氨酸(Glu)的浓度、谷氨酰胺合成酶(GS)酶活也上调。结论:场景恐惧记忆过程中海马区发生了形态学和功能学的神经可塑性改变,其中兴奋性神经递质谷氨酸得到显著上调,可能是由于其谷氨酸—谷氨酰胺代谢偶联的潜在机制导致。Objective:The current study aims to probe and confirm the morphological characteristics and the excitatory neurotransmitters of hippocampus in the contextual fear memory model of post-traumatic stress disorder.In addition,the study exploring the potential mechanism of the changes.Methods:30 C57 male mice were randomly divided into 2 groups,Golgi staining,and detection kits were used to detect the morphological structure,the levels of glutamic acid and the enzyme activity of glutamine synthetase in contextual fear memory model.Results:Quantitative and comparative data show that the density of dendritic spines was higher in the hippocampus area of PTSD mice than control group.At the same time,the levels of glutamic acid(Glu)and the enzyme activity of glutamine synthetase(GS)in contextual fear memory model also significant increased compared to the control group.Conclusion:The present results suggested that the morphological and functional plasticity improved of hippocampus during fear memory.Expecially the excitatory neurotransmitter glutamic acid was significantly up-regulated,which may be caused by the glumate-glutamine metabolic couping.
