摘  要:目的 探讨高强度超声介入治疗对宫颈癌小鼠模型肿瘤增殖、免疫功能及脾细胞Th1/Th2亚群的影响。方法 42只宫颈癌小鼠按随机数字表法分为3组-模型组、超声1组与超声2组,每组14只。超声1组与超声2组均给予进行局部高强度超声治疗,超声1组的单点单次治疗时间为10 s,超声2组的单点单次治疗时间为20 s,共治疗15次;模型组不给予超声治疗。所有小鼠均治疗观察4周,记录3组小鼠的体重、肿瘤体积、肿瘤增殖、免疫功能及脾细胞Th1/Th2亚群变化情况。结果 超声1组与超声2组治疗第2周与第4周的小鼠体重均高于模型组,肿瘤体积均小于模型组,超声2组较超声1组变化更显著,差异均有统计学意义(P<0.05)。超声1组与超声2组治疗第4周与第8周的小鼠肿瘤细胞增殖率均低于模型组,超声2组低于超声1组,差异均有统计学意义(P<0.05)。超声1组与超声2组治疗第4周与第8周的CD4^(+)T细胞比例和CD8^(+)T细胞比例高于模型组,超声2组高于超声1组,差异均有统计学意义(P<0.05)。超声1组与超声2组治疗第4周与第8周的Bcl-2、MMP-9蛋白相对表达水平低于模型组,超声2组低于超声1组,差异均有统计学意义(P<0.05)。结论 高强度超声介入在宫颈癌小鼠的应用能抑制Bcl-2、MMP-9蛋白的表达,改善脾细胞Th1/Th2亚群功能,提高小鼠的免疫功能,抑制肿瘤增殖,从而发挥很好的抗癌作用。Objective To investigate the effects of high Intensity Ultrasound Interventional Therapy on tumor proliferation, immune function and Th1/Th2 subsets of spleen cells in a mouse model of cervical cancer. Methods Forty-two cervical cancer mice were divided into 3 groups by random number table method-model group, ultrasound group 1 and ultrasound group 2, 14 mice in each group. Both ultrasound group 1 and ultrasound group 2 were given local High Intensity Ultrasound Interventional Therapy. The single-point single treatment time of ultrasound group 1 were 10 s, and the single-point single treatment time of ultrasound group 2 were 20 s, with the total of 15 treatments;the model group were not given ultrasound treatment. All mice were treated and observed for 4 weeks, and the body weight, tumor volume, tumor proliferation, immune function, and changes of Th1/Th2 subgroups of spleen cells in the three groups were recorded. Results The mice in the ultrasound group 1 and ultrasound group 2 and the second week and the fourth week of treatment had higher body weights than the model group, and the tumor volume were less than the model group, the change of ultrasound group 2 was more significant than that of ultrasound group 1, the differences were statistically significant(P<0.05). The proliferation of tumor cells in the ultrasound group 1 and ultrasound group 2 was lower than that in the model group at the 4th and 8th week of treatment, and the ultrasound group 2 was lower than the ultrasound group 1, the differences were statistically significant(P<0.05). The ratio of CD4^(+)T cells and CD8^(+)T cells in the 4th and 8th weeks of ultrasound treatment in the ultrasound group 1 and ultrasound group 2 were higher than that of the model group, and ultrasound group 2 were higher than ultrasound group 1, the differences were statistically significant(P<0.05).
