摘 要:为探明北美豆梨和杜梨花粉萌发的适宜条件和最佳液体培养基组分,以北美豆梨‘克利夫兰’‘三体’和我国的杜梨花粉为材料进行了培养温度,蔗糖、H_(3)BO_(3)和Ca(NO_(3))_(2)质量浓度以及贮藏方式对花粉萌发影响的试验,利用L_(16)(4^(3))正交试验设计对液体培养基组分进行了筛选优化。结果表明:杜梨及北美豆梨花粉萌发的适宜温度为25℃,蔗糖质量浓度为150 g·L^(-1);适宜杜梨、‘克利夫兰’和‘三体’花粉萌发的H3BO3质量浓度分别为50,100,50 mg·L^(-1),Ca(NO_(3))_(2)质量浓度分别为50,100,50 mg·L^(-1);-20℃和-40℃冷冻贮藏是3种花粉保存的最佳方式;杜梨、‘克利夫兰’和‘三体’花粉萌发的最适液体培养基分别为:150 g·L^(-1)蔗糖+100 mg·L^(-1)Ca(NO_(3))_(2)+50 mg·L^(-1)H_(3)BO_(3),100 g·L^(-1)蔗糖+100 mg·L^(-1)Ca(NO_(3))_(2)+150 mg·L^(-1)H_(3)BO_(3),130 g·L^(-1)蔗糖+50 mg·L^(-1)Ca(NO_(3))_(2)+150 mg·L^(-1)H3BO3。The effects of culture temperature,sucrose,boric acid and calcium nitrate concentrations and storage methods on pollen germination were studied with Pyrus calleryana and Pyrus betulaefolia pollen as materials to find out the suitable conditions and the best liquid medium components for pollen germination of Pyrus calleryana and Pyrus betulaefolia.The liquid medium components were screened and optimized by L_(16)(4^(3))orthogonal experiments.The results showed that the suitable temperature for pollen germination of Pyrus betulaefolia and Pyrus calleryana was 25℃,and the sucrose concentration was 150 g·L^(-1).The suitable boric acid concentration for pollen germination of Pyrus betulaefolia,‘Cleveland’and‘Trisomy’were 50,100,50 mg·L^(-1),and the suitable calcium nitrate concentration were 50,100,50 mg·L^(-1).-20℃and-40℃frozen storage is the best way to preserve three kinds of pollen;the optimum liquid medium for pollen germination of Pyrus betulaefolia,‘Cleveland’and‘Trisomic’were:150 g·L^(-1)sucrose+100 mg·L^(-1)calcium nitrate+50 mg·L^(-1)boric acid,100 g·L^(-1)sucrose+100 mg·L^(-1)calcium nitrate+150 mg·L^(-1)boric acid,130 g·L^(-1)sucrose+50 mg·L^(-1)calcium nitrate+150 mg·L^(-1)boric acid,respectively.
