摘 要:目的 探讨结直肠癌组织中微小RNA-876-5p(miR-876-5p)、配对框基因6(PAX6) mRNA的表达变化及意义。方法 选取86例结直肠癌患者,术中留取部分癌组织和癌旁组织。用RT-qPCR法检测组织中miR-876-5p、PAX6 mRNA表达;用TargetScan数据库预测miR-876-5p与PAX6的关系,用Pearson相关法分析二者的相关性;随访3年,统计患者3年累积生存率,根据结直肠癌组织中miR-876-5p、PAX6 mRNA表达水平将患者分为miR-876-5p高表达组、miR-876-5p低表达组,PAX6 mRNA高表达组、PAX6 mRNA低表达组,比较各组3年累积生存率。结果 结直肠癌组织中miR-876-5p相对表达量低于癌旁组织,PAX6 mRNA相对表达量高于癌旁组织(P均<0.05)。经在线网站https://www. targetscan. org/预测,miR-876-5p与PAX6的3’-非翻译区691-697存在结合位点。Pearson相关分析显示,结直肠癌组织中miR-876-5p表达与PAX6 mRNA表达呈负相关(r=-0.754,P<0.05)。结直肠癌组织中miR-876-5p、PAX6 mRNA表达与肿瘤分化程度、TNM分期、淋巴结转移有关(P均<0.05),与性别、年龄、肿瘤部位、肿瘤最大径无关(P均>0.05)。miR-876-5p高表达组3年累积生存率90.48%(38/42)高于miR-876-5p低表达组的68.18%(30/44),PAX6 mRNA高表达组3年累积生存率70.21%(33/47)低于PAX6 mRNA低表达组的89.74%(36/39),比较差异均有统计学意义(P均<0.05)。结论 结直肠癌组织中miR-876-5p高表达、PAX6 mRNA低表达,二者表达变化与肿瘤分化程度、TNM分期、淋巴结转移及预后有关。Objective To investigate the expression changes and significance of microRNA-876-5p(miR-876-5p) and paired box 6(PAX6) in colorectal cancer tissues. Methods Eighty-six colorectal cancer patients were selected, and part of the cancerous tissue and paracancerous tissue were retained during the operation. The miR-876-5p and PAX6mRNA expression levels in tissues were detected by RT-qPCR. TargetScan database was used to predict the relationship between miR-876-5p and PAX6, and Pearson correlation method to analyze the correlation between the two indicators. Patients were followed up for 3 years, and we calculated the 3-year cumulative survival rate of patients. According to the expression levels of miR-876-5p and PAX6 mRNA in the colorectal cancer tissues, the patients were divided into the high miR-876-5p expression group, low miR-876-5p expression group, high PAX6 mRNA expression group, and low PAX6 mRNA expression group;we compared the 3-year cumulative survival rate. Results The relative expression of miR-876-5p in the colorectal cancer tissues was lower than that in the adjacent tissues, and the relative expression of PAX6 mRNA was higher than that in the adjacent tissues(both P<0. 05). Pearson correlation analysis showed that there was a negative correlation between miR-876-5p expression and PAX6 mRNA expression in colorectal cancer(r=–0. 754, P<0. 05). The expression levels of miR-876-5p and PAX6 mRNA in the colorectal cancer tissues were related to degree of tumor differentiation, TNM stage, and lymph node metastasis(all P<0. 05), were not related to sex, age, tumor location, or the largest diameter of tumor(all P>0. 05). The 3-year cumulative survival rate of the high miR-876-5p expression group was 90. 48%(38/42), which was higher than that [68. 18%(30/44)]of the low miR-876-5p expression group.
