摘  要:目的 比较Beagle犬牙周韧带成纤维细胞(periodontal ligament fibroblasts cells, PDLFs)和骨髓基质细胞(bone marrow stromal cells, BMSCs)体外成骨潜能,为筛选合适的种子细胞促进牙周组织再生提供实验依据。方法 体外通过茜素红染色、Realtime-PCR比较两种细胞碱性磷酸酶(alkaline phosphatase, ALP)及骨保护素(osteoprotegerin, OPG)的表达。结果 茜素红染色显示PDLFs成骨能力高于BMSCs;Realtime-PCR显示,PDLFs的OPG表达量高于BMSCs,差别有统计学意义(P<0.05);PDLFs的ALP表达量虽低于BMSCs,但两者之间差别无统计学意义(P>0.05)。结论 PDLFs体外成骨潜能不低于BMSCs。Objective To compare the osteopenic potential between periodontal ligament fibroblasts cells(PDLFs) and bone marrow stromal cells(BMSCs) in Beagle dog, and provide foundation for selecting seed cells of periodontal tissue regeneration.Methods Alizarin red staining and realtime PCR were used to compare the expression of alkaline phosphatase(ALP) and osteoprotegerin(OPG) between PDLFs and BMSCs.Results The result of Alizarin red staining showed that the osteopenic potential of PDLFs was higher than that of BMSCs.Date of realtime PCR showed that the levels of OPG in PDLFs was higher than that in BMSCs(P<0.05);the levels of ALP in PDLFs was lower than that in BMSCs, but the result had no statistical significance(P>0.05).Conclusion The osteopenic potential of PDLFs was not lower than that of BMSCs.
