摘 要:目的 通过有效成分测定、自由基清除和免疫调节实验研究植物酵素/蜂王浆混合物的生物活性。方法 采用HPLC法、BCA法及亚硝酸钠-硝酸铝比色法分别测定10-羟基-2-葵烯酸和没食子酸、蛋白质及黄酮类化合物的含量;通过测定植物酵素/蜂王浆混合物对自由基的清除率及免疫低下小鼠免疫指标的影响考察其抗氧化及免疫调节活性。结果 植物酵素/蜂王浆混合物中10-羟基-2-葵烯酸、蛋白质含量分别为3.30 mg·mL^(-1)、145.90 mg·g^(-1),酵素原浆中没食子酸、黄酮类化合物分别为0.35 mg·mL^(-1)、0.16 mg·mL^(-1)。植物酵素/蜂王浆混合物对DPPH·、超氧阴离子自由基及羟自由基的清除率分别为85.11%、72.20%及44.51%。中、高剂量组的植物酵素/蜂王浆混合物均能显著提高模型组小鼠的免疫脏器指数(P <0.05),高剂量组在一定程度上能修复免疫低下小鼠免疫器官的形态结构,植物酵素/蜂王浆混合物各剂量组均可显著提高小鼠血清中IgM、IL-1β和TNF-α细胞因子的含量(P <0.01),具有免疫增强作用。结论 植物酵素/蜂王浆混合物具有较好的抗氧化性能及增强免疫低下小鼠免疫功能的作用。Objective To determine the biological activity of a plant enzyme/royal jelly mixture through active ingredient measurement, free radical scavenging and immunomodulation experiments. Methods The content of 10-hydroxydec-2-enoic acid and gallic acid, protein and flavonoids was determined by HPLC, BCA and sodium nitrite-aluminium nitrate colorimetric methods, respectively. The antioxidant and immunomodulatory activities of the plant enzyme/royal jelly mixture were determined by measuring the scavenging rate of free radicals and the immune indexes in immunocompromised mice. Results The content of 10-hydroxydec-2-enoic acid and protein in the plant enzyme/royal jelly mixture was 3.30mg·mL^(-1)and 145.90 mg·g^(-1), and the content of gallic acid and flavonoids in the plant enzyme original syrup was 0.35 mg·mL^(-1)and 0.16 mg·mL^(-1). The scavenging rates of DPPH·, superoxide anion radicals and hydroxyl radicals by the plant enzyme/royal jelly mixture was 85.11%, 72.20% and 44.51%, respectively. The plant enzyme/royal jelly mixture in the medium and high dose significantly improved the immune organ indexes of mice in the model group(P < 0.05), and the high dose group repaired the morphological structure of immune organs in immunocompromised mice to a certain extent. In addition, all dose groups of the plant enzyme/royal jelly mixture increased the levels of IgM,IL-1β and TNF-α cytokines in the mice serum(P < 0.01), displaying an immune-enhancing effect.Conclusion Plant enzyme/royal jelly mixture has good antioxidant properties and may enhance immune the function in immunocompromised mice.
