摘  要:目的探讨常染色体STR和X染色体STR基因座分型在叔侄关系鉴定中的应用策略。方法对本案例中被检女孩、疑为女孩生母和另外3人(女性1名,疑为被检女孩的姑姑;男性2名,疑为被检女孩的叔叔)血样DNA提取后,分型采用PowerPlex■Fusion、Mircroreader^(TM)23_(SP)ID系统和Microreader^(TM)19X ID系统进行常染色体和X染色体STR复合扩增,毛细管电泳后分析各STR基因型。结果根据常染色体STR分型和ITO法计算出的叔侄姑侄指数,支持姑侄关系,不排除叔侄关系,家系基因型重建法推导的祖父母基因型符合作为被检女孩亲生祖父母的遗传条件,X-STR基因分型支持祖母-孙女关系。结论对于叔侄等复杂亲缘关系鉴定,需要综合运用多种遗传标记(常染色体STR、性染色体STR)和多系统联合分析,互相验证,才有利于获得较为可靠的鉴定意见。Objective To discuss the strategies for the determination of uncle-niece relationship through the detection of autosomal and X-STR loci.Methods STR genotypes were performed by using PowerPlex■Fusion,Mircroreader^(TM)23_(SP)ID systems and Microreader^(TM)19X ID system from five persons,who are the daughter,suspected-mother and suspected-paternal relatives of three uncles and aunts.Results Relationships between the detected uncle/aunt and niece were not excluded based on the results of autosomal STR genotypes and avuncular index(AI),and pedigree genotype reconstruction method analysis and X-STR genotypes also had the same conclusion.Conclusion Combined analysis of multi-genotyping system and multiple methods could help to address more complex kinship tests.
