摘 要:目的 探讨温阳化饮方通过调控mTOR信号通路干预支气管哮喘寒饮蕴肺证大鼠气道炎症的作用机制。方法 采用卵清白蛋白(OVA)致敏,利用冰箱冰冻给予寒冷刺激、冰水喂养给予饮冷刺激以及让大鼠在水中游泳直至其即将沉入水中、难以上浮使其劳累等方法,构建支气管哮喘寒饮蕴肺证的病证结合大鼠模型,并分别给予雷帕霉素、小青龙汤、温阳化饮方进行治疗,观测相关指标的表达情况。结果 模型组大鼠体重较其他组明显降低,支气管黏膜上皮细胞可见严重肿胀、增生、坏死、脱落,肺泡内大量炎性细胞浸润;TGF-β、TNF-α的表达显著增高;Atg 5、12 mRNA的表达明显降低;自噬标志蛋白LC3-Ⅰ向LC3-Ⅱ转化增加,mTOR通路的表达显著降低,Beclin1表达显著增高。而与模型组相比各给药组对上述各项指标有明显的改善,其中温阳化饮方疗效总体最佳。结论 温阳化饮方在支气管哮喘寒饮蕴肺证的治疗中确有较好的治疗效果,其疗效机制可能在于抑制肺组织自噬相关mTOR信号通路的表达,增加自噬基因Atg的表达,以提高自噬活性,减轻气道炎症水平。Objective To explore the mechanism of wenyanghuayin Recipe on airway inflammation in rats with bronchial asthma cold Yin accumulating lung syndrome by regulating mTOR signal pathway.Methods Using ova sensitization,cold stimulation by freezing in the refrigerator,cold stimulation by feeding in ice water,and swimming in the water until the rats are about to sink into the water and are difficult to float up to make them tired,the disease syndrome combined rat model of cold Yin accumulating lung syndrome of bronchial asthma was established,and rapamycin,Xiaoqinglong decoction The expression of related indexes was observed.Results The body weight of the model group was significantly lower than that of other groups.Severe swelling,proliferation,necrosis and abscission of bronchial mucosal epithelial cells were observed,and a large number of inflammatory cells were infiltrated in the alveoli;TGF-β、 TNF-α The expression of was significantly increased;The expression of ATG 5 and 12 mRNA decreased significantly;The transformation of autophagy marker protein LC3-Ⅰ to LC3-Ⅱ increased,the expression of mTOR pathway decreased significantly,and the expression of Beclin1 increased significantly.Compared with the model group,each administration group has significantly improved the above indicators,and the curative effect of wenyanghuayin formula is the best overall.Conclusion Wenyanghuayin decoction has a good therapeutic effect in the treatment of cold Yin accumulating lung syndrome of bronchial asthma.Its therapeutic mechanism may lie in inhibiting the expression of autophagy related mTOR signal pathway in lung tissue and increasing the expression of autophagy gene ATG,so as to improve autophagy activity and reduce the level of airway inflammation.
