摘 要:目的优化菊花明目方提取、干燥工艺,研究其提取物体外抗氧化活性。方法以绿原酸、木犀草苷转移率,出膏率为指标,在单因素试验的基础上结合正交试验优化醇提工艺;比较不同的干燥方法,通过吸湿曲线考察不同辅料对真空干燥效果的影响;并以抗坏血酸(Vc)为阳性对照,采用ABTS、DPPH法对菊花明目方提取物体外抗氧化活性进行了研究。结果最佳提取工艺为用15倍量体积分数70%的乙醇,提取2次,每次提取1.5 h,绿原酸与木犀草苷的转移率分别为97.52%、93.64%,浸膏收得率为36.18%;选择D-甘露糖醇辅助醇提物进行真空干燥;抗氧化活性研究表明,菊花明目方提取物对DPPH自由基、ABTS自由基清除率分别可达(99.75±0.30)%、(44.65±0.13)%。结论优选的提取、干燥工艺稳定,重现性良好。菊花明目方提取物具有良好的抗氧化作用。Objective The extraction and drying process of Chrysanthemum Eyesight prescription were optimized and its antioxidant activity in vitro was studied.Methods The ethanol extraction process was optimized with the transfer rate of chlorogenic acid and luteolin glycoside and the yield of extract as indexes.The effects of different excipients on vacuum drying were investigated by comparing different drying methods and hygroscopic curves.With Vc as positive control,the antioxidant activity of extract of Chrysanthemum Mingmu formula was studied by ABTS and DPPH methods.Results The optimal extraction conditions were as follows:using 15 times of 70% ethanol,extracting twice for 1.5 h each,the transfer rates of chlorogenic acid and luteolin were 97.52% and 93.64%,respectively,and the extract yield was 36.18%.The ethanol extract was vacuum-dried with D-mannitol.The results showed that the scavenging rate of DPPH free radical and ABTS free radical was(99.75±0.30)% and(44.65±0.13)%,respectively.Conclusion The optimized extraction and drying process was stable and reproducible.The extract of Chrysanthemum Eyesight prescription has good antioxidant effect.
