番茄红素对限制应激着床期孕鼠子宫内膜发育和胚胎着床的影响

摘  要:试验旨在探究番茄红素(LYC)对限制应激孕鼠着床期子宫内膜发育和胚胎着床数目的影响。选择8周龄昆明系孕鼠80只,在妊娠第1 d随机分为4组,每组3个重复,每个重复10只,分别为对照组、限制应激组(RS组)、玉米油+应激组(CRS组)、LYC+应激组(LRS组)。将LYC按10 mg/kg (体重)剂量给孕鼠灌胃(0.5 mL/只),灌胃2 h后进行限制应激处理(4 h/d),连续处理5 d,即胚胎着床期第5 d时,取血清和子宫组织。结果显示,与RS组相比,LRS组孕鼠血清中去甲肾上腺素含量降低10.00%,胚胎着床数增加47.92%,子宫指数增加83.36%(P<0.05),子宫内膜结构趋向对照组,表现为子宫内膜疏松,在靠近子宫腔附近细胞出现明显蜕膜化,子宫腺数量增多。进一步经荧光定量PCR和分光光度法检测发现,LYC预处理能够显著增加应激孕鼠子宫核因子红细胞相关因子2 (Nrf2) mRNA表达,改变Nrf2下游信号抗氧化酶水平。与RS组相比,LRS组着床期子宫中超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)活性分别增加21.10%、82.60%,总抗氧化能力(T-AOC)显著增加64.50%,而丙二醛(MDA)含量显著降低65.60%(P<0.05)。研究表明,LYC可增加限制应激孕鼠着床期子宫中Nrf2表达,降低子宫局部氧化应激水平,提高子宫局部免疫水平,进而缓解应激对子宫内膜蜕膜发育的不利影响,增加胚胎着床数目。The study was to investigate the effect of lycopene(LYC) on endometrial development and embryo implantation number in stressed pregnant mice. A total of 80 eight-week-old Kunming pregnant mice were randomly divided into four groups on the 1st day of gestation, with three replicates in each group and ten mice in each replicate:control group, restricted stress group(RS group), corn oil + stress group(CRS group) and LYC+ stress group(LRS group). Pregnant rats were given LYC at the dose of 10 mg/kg(body weight) by intragastrictive administration(0.5 mL/piece). After 2 h of intragastrictive administration, restricted stress treatment(4 h/d) was carried out for five consecutive days, and serum and uterine tissue were collected at fifth day of embryo implantation stage. The results showed that compared with the RS group, the serum norepinephrine content of pregnant mice in LRS group was significantly decreased by 10.00%, the number of embryo implantation was increased by 47.92%, and the uterine index was increased by 83.36%(P<0.05). The endometrial structure of pregnant mice in the LRS group tended to be that of control group, and the endometrial structure showed loose endometrium, obvious decidualization of cells near the uterine cavity, and the number of uterine glands increased. Further detection by fluorescence quantitative PCR and spectrophotometry showed that LYC pretreatment could significantly increase the expression of Nrf2 mRNA in the uterus of stressed pregnant rats, and change the level of Nrf2 downstream signaling antioxidant enzymes. Compared with RS group, the activity of SOD and GSH-Px in uterus during implantation in LRS group were increased by 21.10% and 82.60%, respectively, and T-AOC was significantly increased by 64.50%. The content of MDA was significantly decreased by 65.60%(P<0.05). The experiment indicates that LYC can increase the expression of Nrf2 in uterus of stressed pregnant rats during implantation, reduce the level of local oxidative stress in the uterus, improve the level of local immun
