摘　　要：吉式库特菌在自然界普遍存在,有的作为机会致病菌可引起动物疾病。本研究发现了1株吉式库特菌(Kurthia gibsonii)噬菌体vb_KgiM_P1(简称MP1),并对其生物学特性和全基因组进行研究。透射电镜观察显示MP1具有典型的二十面体结构的头部和可收缩的尾部,属于肌尾病毒科(Myoviridae)。生物学特性研究表明,MP1的最佳感染复数为1,裂解周期在50 min左右,潜伏期约为15 min,暴发期约为50 min,平均暴发量约为83 PFU/cell,表明该噬菌体有高效的杀菌活性。MP1的稳定性良好,在25~50℃之间和pH值3~11之间保持稳定的裂解活性。全基因组测序显示该噬菌体基因组全长为15855 bp,G+C含量为32%,有20个开放阅读。系统发育分析结果表明MP1与葡萄球菌噬菌体和肠球菌噬菌体属于共同起源的不同进化分支,且相似性较低,证明MP1是1株全新的烈性噬菌体。Kurthia gibsonii is ubiquitous in nature and some act as opportunistic pathogenic bacteria that can cause disease in animals.This study aims to study the biological characteristics of a phage of Kurthia gibsonii and analyze the whole genome of the phage.A phage of Kurthia gibsonii was isolated and named vb-KgiM-P1,or MP1 for short.Electron microscopy showed that MP1 had a head with a typical icosahedral structure and a retractable tail,belonging to the family Myoviridae.Biological characteristics studies showed that the optimal multiplicity of infection for MP1 was 1,the lysis cycle was 50 min,the latent period was about 15 min,the burst period was about 50 min,and the average burst size was about 83 PFU/cell.MP1 was stable and had lytic activity at 25℃to 50℃and pH3-pH11.Whole-genome sequencing indicated that the full length of the phage was 15855 bp and the G+C content was 32%.MP1 had 20 open reading frames.The results of the phylogenetic analysis showed that MP1 and Staphylococcus phages and Enterococcus phages belonged to different evolutionary branches of common origin.The similarity was low,proving that MP1 was completely new.
