摘 要:目的 利用iTRAQ标记的蛋白组学技术探讨骨肉瘤肺转移患儿血清中蛋白的表达。方法 纳入10例骨肉瘤患儿和10例骨肉瘤肺转移患儿作为研究对象,收集所有研究对象血清5 ml,采用iTRAQ标记的蛋白组学技术分析2组血清中的差异表达蛋白,并对差异表达蛋白进行GO富集分析、KEGG通路富集及蛋白-蛋白互作分析。外科手术后获取2例骨肉瘤肺转移患儿及2例骨肉瘤患儿瘤体组织,采用Western blot验证2组患儿癌组织中CDK1、hnRNP A2/B1、ITGAV和SRSF10的蛋白表达水平。结果 与骨肉瘤患儿相比,骨肉瘤肺转移患儿血清中高表达的蛋白共477种,低表达的蛋白共235种。KEGG通路富集显示,ECM及受体的相互作用、黏着力及PI3K-Akt信号通路等可能是骨肉瘤肺转移的主要发病机制。骨肉瘤肺转移患儿血清中差异蛋白前10种为CDK1、CTNNB1、ITGB1、SNRPE、SNRPG、HNRNPH1、SNRPD3、HNRNPC、ITGAV和SRSF1。与骨肉瘤患儿的癌组织相比,骨肉瘤肺转移患儿癌组织中CDK1、hnRNP A2/B1、ITGAV和SRSF10蛋白表达水平升高(P<0.05)。结论 与骨肉瘤患儿相比,骨肉瘤肺转移患儿血清中存在多种差异表达蛋白,采用iTRAQ标记的蛋白组学技术能很好地筛选出差异蛋白,为骨肉瘤发生肺转移的分子机制研究提供了潜在的血清学标志物。Objective This paper aims to investigate the serum protein expression level in children with lung metastases of osteosarcoma via iTRAQ-labeled proteomics technique. Methods A total of 10 children with osteosarcoma and 10 children with lung metastasis of osteosarcoma were enrolled. About 5 ml of serum from all children were collected. The iTRAQ-labeled proteomics technique was used to analyze the differentially expressed proteins in the serum of the two groups. The differential expressed proteins were further analyzed via GO enrichment analysis, KEGG pathway enrichment and protein-protein interaction analysis. After surgery, tumor tissues from 2 children of lung metastasis of osteosarcoma and 2 children of osteosarcoma were obtained. Western blot was used to verify the protein expression levels of CDK1, hnRNP A2/B1, ITGAV and SRSF10 in the two groups. Results Compared with osteosarcoma, 477 up-regulated proteins and 235 down-regulated proteins were observed in the serum of children with lung metastases of osteosarcoma. The KEGG pathways shows that the interaction between ECM and receptor, focal adhesion and PI3K-Akt signaling pathway may be the main pathogenesis of lung metastasis of osteosarcoma. The top 10 differential expressed proteins in serum of children with lung metastasis of osteosarcoma include CDK1, CTNNB1, ITGB1, SNRPE, SNRPG, HNRNPH1, SNRPD3, HNRNPC, ITGAV and SRSF1. Compared with osteosarcoma, the protein levels of CDK1, hnRNP A2/B1, ITGAV and SRSF10 in lung metastasis of osteosarcoma were increased(P<0.05). Conclusion Compared with osteosarcoma, there are a variety of differentially expressed proteins in the serum of children with lung metastasis of osteosarcoma. iTRAQ-labeled proteomics technique can screen the differential expressed proteins, which provides potential serological markers in the molecular mechanism of lung metastasis of osteosarcoma.
