肝細胞癌ZHX2蛋白表達及其與臨床病理特徵的關係

摘　　要：目的探討肝細胞癌(hepatocellular carcinoma,HCC)、癌旁肝組織和肝血管瘤旁正常肝組織中鋅指和同源盒2(zinc-fingers and homeoboxes 2,ZHX2)蛋白的表達及分析其與臨床病理特徵之間的關係。方法收集肝臟手術標本,包括HCC(其中含癌旁肝組織)和血管瘤旁正常肝組織,並製作組織芯片。ZHX2蛋白表達採用免疫組織化學法檢測,分析ZHX2與臨床病理特徵的關係。結果ZHX2在HCC中呈現不同程度陽性,陽性表達率為37.1%(117/315);癌旁肝組織陽性表達率為49.2%(90/183例);血管瘤旁正常肝組織陽性表達率為58.6%(58/99)。HCC ZHX2陽性表達率明顯低於癌旁肝組織和血管瘤旁正常肝組織(P<0.05);AFP水平正常的HCC患者ZHX2表達高於AFP水平異常的HCC患者(P<0.05),ZHX2表達與其它臨床病理特徵無相關性。結論轉錄抑制因子ZHX2的低表達可能參與HCC的發生發展,ZHX2免疫組化檢測可以作為血清AFP水平正常的HCC患者的補充診斷指標。Objective To explore the expression of zinc-fingers and homeoboxes 2(ZHX2)protein in hepatocellular carcinoma(HCC),paracancerous liver tissue,and normal liver tissue adjacent to hemangiomas,and analyze its correlation with clinicopathological features.Methods Liver surgery specimens including HCC,paracancerous liver tissue and normal liver tissue adjacent to hemangiomas were collected and made tissue microarray.The expression of ZHX2 protein was detected by immunohistochemistry.The relationships between ZHX2 and clinopathological features were analyzed.Results ZHX2 showed various degrees of response and the positive rate was 37.1%(117/315)in HCC.ZHX2 positive rate was 49.2%(90/183)in paracancerous liver tissue and 58.6%(58/99)in normal liver tissue adjacent to hemangiomas.The positive rate of ZHX2 in HCC was significantly lower than that in paracancerous liver tissue and normal liver tissue adjacent to hemangiomas(P<0.05).The expression of ZHX2 in HCC patients with normal AFP levels was higher than that in HCC patients with abnormal AFP levels(P<0.05).The expression of ZHX2 did not correlate with other clinicopathological features.Conclusion Low expression of transcription repressor ZHX2 may involve in carcinogenesis and progress of HCC.Immunohistochemistry of ZHX2 can be used as a supplementary indicator for HCC patients with normal serum AFP levels in diagnosis.
