摘  要:目的探讨miR-100-5p靶向mTOR对急性髓样白血病细胞增殖的影响。方法miR-100-5pmimics转染HL-60细胞,通过实时定量PCR进行验证及Western blot检测miR-100-5p过表达后细胞mTOR蛋白的表达。采用CCK-8法检测miR-100-5p对HL-60细胞增殖的影响,应用双荧光素酶报告基因实验进行验证mTOR是否为miR-100-5p潜在的靶基因,CCK-8法进一步检测miR-100-5p靶向mTOR对急性髓样白血病细胞增殖的影响。结果miR-100-5p下调HL-60细胞中mTOR mRNA和蛋白表达水平,抑制HL-60细胞体外增殖,mTOR为miR-100-5p的靶标,且miR-100-5p靶向mTOR抑制急性髓样白血病细胞增殖。结论miR-100-5p通过靶向mTOR抑制急性髓样白血病中的细胞增殖。Objective To explore the effect of miR-100-5p targeting m TOR on the proliferation of acute myeloid leukemia cells.Methods HL-60 cells were transfected with miR-100-5p mimics,and the expression of mTOR mRNA and protein was detected by real-time quantitative PCR and Western blot.The effect of miR-100-5p on the proliferation of HL-60 cells was detected by CCK-8 method,and the double luciferase reporter gene experiment was used to verify whether mTOR was a potential target gene of miR-100-5p.The effect of miR-100-5p targeting m TOR on the proliferation of acute myeloid leukemia cells was further detected by CCK-8 method.Results miR-100-5p down-regulated the expression of mTOR mRNA and protein in HL-60 cells and inhibited the proliferation of HL-60 cells.mTOR was the target of miR-100-5p,and miR-100-5p targeted mTOR to inhibit the proliferation of acute myeloid leukemia cells.Conclusion miR-100-5p inhibits the proliferation of acute myeloid leukemia cells via targeting mTOR.
