摘 要:目的探索过表达miR-145对卵巢癌细胞增殖和侵袭的影响及可能机制。方法通过qRT-PCR检测miR-145和SOX2在卵巢癌细胞中的表达,体外培养人卵巢癌SKOV3细胞,转染miR-145模拟物,MTT法检测过表达miR-145对SKOV3细胞增殖的影响,Transwell实验检测过表达miR-145对SKOV3细胞侵袭能力的影响。应用qRT-PCR和Western blot检测过表达miR-145对SOX2表达的影响,生物学网站分析及双荧光素酶报告基因实验研究miR-145和SOX2的靶向关系。结果miR-145在卵巢癌细胞中低表达,SOX2在卵巢癌细胞中高表达,转染miR-145模拟物可以增加卵巢癌细胞中miR-145的表达,降低SOX2的表达,且在SOX2基因3’UTR区存在miR-145的结合位点,双荧光素酶报告基因实验显示SOX2是miR-145的靶基因。过表达miR-145可以抑制卵巢癌细胞的增殖及侵袭。结论过表达miR-145可以抑制卵巢癌细胞SKOV3的增殖及侵袭能力,机制可能与靶向负调控SOX2的表达有关。Objective To explore the effect and possible mechanism of overexpression of miR-145 on proliferation and invasion of ovarian cancer cells.Methods The expressions of miR-145 and SOX2 in human ovarian cancer cell line and normal human ovarian epithelial cell line were detected by qRT-PCR.The miR-145 mimics was transfected into SKOV3 cells to upregulate miR-145 expression.The effect of overexpression of miR-145 on the proliferation of SKOV3 cells was detected by MTT assay.The effect of overexpression of miR-145 on the invasion ability of SKOV3 cells was detected by Transwell assay.qRT-PCR and Western blot were used to detect the influence of overexpressed miR-145 on the expression of SOX2.The targeting relationship between miR-145 and SOX2 was studied by biological website analysis and double luciferase reporter assay.Results The expression of miR-145 was low in ovarian cancer cells,while the expression of SOX2 was high in ovarian cancer cells.Transfection of miR-145 mimicry could increase the expression of miR-145 and decrease the expression of SOX2.There was a binding site of miR-145 in the 3’UTR region of SOX2 gene.Dual luciferase reporter assay showed that SOX2 was the target gene of miR-145.Overexpression of miR-145can inhibit the proliferation and invasion of ovarian cancer cells.Conclusion Overexpression of miR-145 can inhibit the proliferation and invasion of SKOV3 in ovarian cancer cells,and the mechanism may be related to the targeted negative regulation expression of SOX2.
