摘 要:目的探究虾青素对心肌梗死后心衰大鼠心肌细胞内皮间充质转化的作用及可能机制。方法40只SD大鼠随机分为假手术组(Sham组)、模型组(Model组)、虾青素低剂量组(AST-L组)和虾青素高剂量组(AST-H组),每组10只。生物信号记录仪检测大鼠血流动力学参数;HE染色观察大鼠心肌病理学改变;Masson染色检测大鼠心肌组织纤维化;荧光探针检测大鼠心肌组织活性氧水平;Western blot检测大鼠心肌组织CD31、VE-cadherin、α-SMA、Vimentin、TGF-β1、Smad3和Smad7蛋白表达水平;qRT-PCR检测大鼠心肌组织TGF-β1、Smad3和Smad7 mRNA表达水平。结果虾青素改善心衰大鼠心功能和心肌组织病理损伤,降低心肌组织纤维化和ROS水平,抑制心肌组织内皮间充质转化,降低TGF-β1和Smad3表达,增加Smad7表达。结论虾青素通过调控TGF-β1/Smad3/7信号通路改善心肌梗死所致心衰大鼠心肌损伤。Objective To explore the effect and possible mechanism of astaxanthin on endothelial mesenchymal transformation of myocardial cells in rats with heart failure after myocardial infarction.Methods Forty SD rats were randomly divided into sham group,model group,AST-L group and AST-H group,with 10 rats in each group.Biological signal recorder was used to detect hemodynamic parameters of rats.HE staining was used to observe pathological changes of rat myocardium.Masson staining was used to detect myocardial fibrosis.Fluorescence probe was used to detect reactive oxygen species in rat myocardial tissue.Western blot was used to detect the expression of CD31,VE-cadherin,α-SMA,Vimentin,TGF-β1,Smad3 and Smad7,and qRT-PCR was used to detect the level of TGF-β1,Smad3 and Smad7 mRNA.Results Astaxanthin improved cardiac function and pathological damage of myocardium in rats with heart failure,decreased fibrosis and ROS levels,inhibited endothelial mesenchymal transformation of myocardium,decreased expression of TGF-β1 and Smad3,and increased expression of Smad7.Conclusion Astaxanthin can improve myocardial injury in heart failure rats induced by myocardial infarction by regulating TGF-β1/Smad3/7 signaling pathway.
