百里香醌对高血脂症心肌损伤保护作用的研究

摘  要:目的 探讨百里香醌对高脂饮食诱导的低密度脂蛋白受体基因敲除(LDLR^(-/-))小鼠心肌损伤的保护作用及机制。方法 选取24只8周龄雄性LDLR^(-/-)小鼠,随机分为正常组、高脂组、高脂+百里香醌组,每组8只。正常组给予普通饮食,高脂组、高脂+百里香醌组给予高脂饮食,高脂+百里香醌组同时给予百里香醌灌胃,3组均干预8周。记录小鼠体重,检测小鼠血清总胆固醇(TC)、低密度脂蛋白胆固醇(LDL-C)、超敏C反应蛋白(hs-CRP)水平,HE染色及Masson染色观察小鼠心肌组织病理形态,免疫组织化学法观察小鼠心肌组织中清道夫受体(SR)、CD36、CD68、过氧化物酶体增殖物激活受体α(PPAR-α)、核因子E2相关因子2(Nrf2)、血红素加氧酶-1(HO-1)、Smad同源物3(Smad3)的表达情况。结果 干预结束后,3组小鼠体重比较差异均无统计学意义(P均>0.05);高脂组血清TC、LDL-C、hs-CRP水平均明显高于正常组(P均<0.05),高脂+百里香醌组血清TC、LDL-C、hs-CRP水平均明显低于高脂组(P均<0.05)。HE染色及Masson染色显示高脂组小鼠心肌组织存在明显炎症细胞浸润和心肌纤维化;高脂+百里香醌组小鼠心肌炎症细胞浸润较少,心肌纤维化程度较轻。免疫组织化学检测显示高脂组小鼠心肌组织中SR、CD36、CD68、Smad3阳性率均明显高于正常组(P均<0.05),高脂+百里香醌组均明显低于高脂组(P均<0.05);高脂组小鼠心肌组织中PPAR-α、Nrf2、HO-1阳性率均明显低于正常组(P均<0.05),高脂+百里香醌组均明显高于高脂组(P均<0.05)。结论 百里香醌可通过减少脂质沉积及抑制氧化应激反应而减轻高血脂症所致心肌损伤。Objective It is to investigate the protective effect and mechanism of thymoquinone on myocardial injury induced by high-fat diet in low-density lipoprotein receptor gene knockout(LDLR^(-/-)) mice. Methods Eight-week-old male LDLR^(-/-)mice were selected and randomly divided into normal diet group, high-fat diet group and high-fat diet+thymoquinone group, each group had 8 mice. The normal group was given a normal diet, the high-fat group and the high-fat+thymoquinone group were given a high-fat diet, and the high-fat+thymoquinone group was given thymoquinone by gavage at the same time, and all three groups were treated for 8 weeks. The body weight of the mice was recorded, and the levels of serum total cholesterol(TC), low-density lipoprotein cholesterol(LDL-C) and hypersensitive C-reactive protein(hs-CRP) were measured. The histopathological morphology of myocardium in the mice was observed by HE staining and Masson staining, the expression of scavenger receptor(SR), CD36, CD68, peroxisome proliferator-activated receptor alpha(PPAR-α), nuclear factor E2-related factor 2(Nrf2), heme oxygenase-1(HO-1), and Smad homolog 3(Smad3) in myocardial tissue of the mice were observed by immunohistochemistry. Results At the end of the intervention, there was no statistically significant difference in body weight among the three groups(all P>0.05);The levels of serum TC, LDL-C and hs-CRP in the high-fat group were significantly higher than those in the normal group(all P<0.05), and the levels of serum TC, LDL-C and hs-CRP in the high-fat+thymoquinone group were significantly lower than those in the high-fat group(all P<0.05). HE staining and Masson staining showed that there were obvious inflammatory cell infiltration and myocardial fibrosis in the myocardial tissues of mice in the high-fat group, the myocardial inflammatory cell infiltration was less and the myocardial fibrosis was less severe in the high-fat+thymoquinone group.
