摘  要:【目的】为高效制备黑果腺肋花楸果实中的抗氧化活性物质,评价其抗氧化活性,提高其利用率和产品附加值,获得与传统提取方式相比更加环保且活性优良的提取方法。【方法】以黑果腺肋花楸果为原料,以ABTS+·和OH·清除率为考察指标,采用响应面法优化超声辅助双水相法萃取黑果腺肋花楸抗氧化成分的工艺,以VC为阳性对照,分别测定了黑果腺肋花楸提取物对OH·、ABTS+·和DPPH·清除率的IC50值,并分析提取物中多酚、黄酮、花青素与抗氧化的相关性。【结果】黑果腺肋花楸抗氧化成分的最佳提取工艺条件为:料液比1∶21 g/mL,硫酸铵质量浓度0.31 g/mL,乙醇体积分数43%,超声功率510 W,超声时间50 min;此时提取物对2种自由基平均清除率为(59.36±0.80)%,其中OH·清除率为(51.77±0.97)%,ABTS+·清除率为(65.20±1.43)%,较单一双水相法和常规醇提法均有显著提升。在此条件下,黑果腺肋花楸提取物得率为(12.67±0.43)%。提取物中多酚、黄酮、花青素含量分别达到了(128.75±0.85)、(47.70±0.74)、(19.95±0.67)mg/g。提取物对ABTS+·、OH·和DPPH·清除率的IC50值分别为0.614、2.273、0.386 mg/mL。【结论】超声辅助双水相法较超声辅助醇提法的ABTS+·清除率提高了8.24%(P<0.01),OH·清除率提高了11.38%(P<0.01);较单一双水相法的ABTS+·清除率提高了10.43%(P<0.01),OH·清除率提高了11.75%(P<0.01)。超声辅助双水相提取法能高效制备黑果腺肋花楸中活性成分,并保持其良好的抗氧化活性。【Objective】This research aimed to efficiently extract the antioxidant components from Aroni amelanocarpa,improve the utilization rate and product-added value of Aroni amelanocarpa,and obtain a more environmentally friendly and active extraction method compared with the traditional extraction method.【Method】Ultrasound technology was used to assist the aqueous twophase extraction of the active constituents.Based on the single-factor experiment,the main process parameters were optimized by the response surface method based on the OH·and ABTS+·radical scavenging capacity.Using VC as a positive control,the IC50values of the radical scavenging capacity for OH·,ABTS+·and DPPH·of Aroni amelanocarpa were measured and the correlation between the main component and the antioxidant index was determined.【Result】The best extraction processing parameter was ascertained as follows:a ratio of material to liquid as 1:21(g/mL),the concentration of ammonium sulfate as 0.31 g/mL,an ethanol concentration of 43%,ultrasonic power as 510 W and extraction time as 50 min.Under these conditions,the average scavenging rate was(59.36±0.80)%,the OH·radical scavenging capacity was(51.77±0.97)%,and the ABTS+·radical scavenging capacity was(65.20±1.43)%.Under these conditions,the yield was(12.67±0.43)%,and the contents of polyphenols,flavonoids and anthocyanins in the extract were(128.75±0.85),(47.70±0.74)and(19.95±0.67)mg/g,respectively.The IC50 values of ABTS,OH and DPPH radical scavenging capacities were 0.614 mg/mL,2.273 mg/mL and 0.386 mg/mL.【Conclusion】Compared with the ultrasonic-assisted ethanol extraction method,the ABTS+·and OH·scavenging capacities were increased by 8.24%(P<0.01)and 11.38%(P<0.01),respectively,by using the ultrasonic-assisted aqueous two-phase extraction method.Compared with the single aqueous two-phase method,the ABTS+and OH·scavenging rates were increased by 10.43%(P<0.01)and 11.75%(P<0.01),respectively,by using the ultrasonic-assisted aqueous two-phase extraction method.Taking advant
