摘　　要：目的研究磷酸化组蛋白H3(phosphorylated histone H3,PHH3)与增殖细胞核蛋白Ki67在胃癌中的表达及其与患者预后的关系,寻找影响胃癌增殖和预后判断的标志物。方法采用石蜡切片免疫组化EnVision染色法检测随访资料完整的70例胃癌及对应癌旁组织中PHH3和Ki67蛋白的表达,分析PHH3和Ki67表达与患者临床病理特征的关系及两者表达的相关性;采用单因素和多因素回归分析影响胃癌患者术后生存率的相关性因素。结果胃癌组织的PHH3和Ki67高表达率高于癌旁组织(P<0.001),PHH3和Ki67在低分化、浸润深度T3~T4和有淋巴结转移的患者中表达高于中高分化、浸润深度T1~T2和无淋巴结转移的患者(P<0.05)。结论PHH3表达上调可能参与了胃癌的发生与发展,并与侵袭、转移等恶性进展有关。Objective This study aims to investigate the expression of phosphorylated histone H3(PHH3)and protein Ki67 of the proliferating nuclear in gastric cancer and their relationship with the prognosis of patients,so as to find the markers that affect the proliferation and prognosis of gastric cancer.Methods Immunohistochemical EnVision staining of paraffin sections was adopted to detect the the expressions of PHH3 and Ki67 in corresponding adjacent tissues of 70 cases of gastric cancer with complete follow-up data.This study analyzed the relation of the expressions of PHH3 and Ki67 with the clinicopathological features of patients,as well as the correlation between the expressions of of PHH3 and Ki67.Univariate and multivariate regression were used to analyze the relevant factors affecting the postoperative survival rate of patients with gastric cancer.Results The high expression rates of PHH3 and Ki67 in gastric cancer tissues were significantly higher than those in paracancer tissues(P<0.001).The expressions of PHH3 and Ki67 were higher in patients with low differentiation,invasion depth of T3~T4 and lymph node metastasis than in those with medium and high differentiation,invasion depth of T1~T2 and no lymph node metastasis(P<0.05).Conclusion The up-regulated expression of PHH3 may be involved in the occurrence and development of gastric cancer,and is related to the malignant progression of invasion and metastasis.
