摘  要:目的 探索在小型猪重组牙胚同种异体移植实现颌骨内牙再生过程中,间充质干细胞BMMSCs的免疫调节作用。方法 将胚胎小型猪帽状期牙胚解离重组后,移植至成年小型猪拔牙区,BMMSCs经分离鉴定后,单独或与他克莫司FK506联合应用,进行同种异体移植免疫干预;评估受体小型猪的免疫状况和牙再生小型猪百分比。结果 BMMSC组受体小型猪CD3^(+)细胞和CD3^(+)CD8^(+)细胞比例均显著下降(P<0.05),Tregs细胞比例显著上调(P<0.05),有牙再生的小型猪百分比为75%;而BMMSC^(+)FK506组CD3^(+)和CD8^(+)T细胞比例均显著下调(P<0.05),但Tregs比例与空白对照组无显著差异,该组25%小型猪有牙再生;空白对照组的免疫排斥相关指标上调,未发现牙再生。结论 相对于与FK506合用,单独使用BMMSCs更有利于同种异体重组牙胚移植。Objective To explore whether MSCs can be applied to reduce immune rejection in allogenic tooth germ transplantation for tooth regeneration in Sus scrofa.Methods In the present study,the allogeneic re-associated tooth germs at cap stage were transplanted into the jawbones of host minipigs.After isolation and identification,allogeneic bone marrow mesenchymal stem cells (BMMSCs) alone or with FK506 were administrated to reduce immune rejection.Immune condition of the host minipigs was evaluated and numbers of minipigs with successfully regenerative teeth were counted.Results Immunological assessment of host minipigs suggested that percentage of CD3^(+)T or CD3^(+)CD8^(+)T cells significantly decreased,whereas that of CD25^(+)FoxP3^(+)Tregs exhibited a significant increase in the peripheral blood after administration of BMMSCs,and the percentage of minipigs with regenerative teeth was 75% in BMMSC group.Allogeneic re-associated tooth germs were all rejected in the negative control group,without tooth regeneration.Conclusions BMMSCs are more beneficial for allogenic re-associated tooth germ transplantation to regenerate tooth in Sus scrofa.
