茶树精油联合云南白药治疗实验性口腔溃疡的疗效及机制研究

摘　　要：目的探究茶树精油联合云南白药治疗治疗实验性口腔溃疡的疗效及机制,为口腔溃疡的临床治疗提供一定的理论依据。方法将48只SD大鼠,随机分成四组,每组12只,分别命名为空白对照组、模型对照组、云南白药组、联合组,采用化学灼烧法建立大鼠口腔溃疡模型,游标卡尺测量创面直径,酶联免疫吸附试验(ELISA)检测各组大鼠血清炎症因子TNF-α、IL-6、IL-2水平,流式细胞术检测各组大鼠外周血T细胞亚群CD4^(+)、CD8^(+)水平。结果茶树精油联合云南白药可以显著缩小大鼠口腔溃疡直径(P<0.05),对模型大鼠口腔溃疡有一定的免疫调节作用,茶树精油和云南白药都能使升高的炎症因子TNF-α、IL-6、IL-2得到降低,使降低的T细胞CD4^(+)、CD4^(+)/CD8^(+)得到回升,升高的CD8^(+)降低,但与单一云南白药治疗相比,茶树精油联合云南白药的免疫调节作用差异显著(P<0.05)。结论茶树精油联合云南白药治疗实验性口腔溃疡可增强疗效,减小溃疡直径,抑制炎症反应,改善免疫调节功能,其治疗机制可能与降低TNF-α、IL-6、IL-2水平和调节CD4^(+)、CD8^(+)T细胞失衡有关。Objective To investigate the effect and mechanism of tea tree essential oil combined with Yunnan Baiyao in the treatment of experimentally oral ulcer.Methods A total of 48 SD rats were randomly divided into four groups of 12,blank control group,model control group,Yunnan baiyao group,combined group.Chemical burning method was adopted to establish the rats oral ulcer model.The levels of serum inflammatory cytokines TNF-α,IL-6 and IL-2 were detected by enzyme-linked immunosorbent assay (ELISA)),and the levels of CD4^(+)and CD8^(+)in peripheral blood T cell subsets were examined by flow cytometry.Results Tea tree essential oil combined with Yunnan baiyao significantly reduced rats oral ulcer diameter (P<0.05).Tea tree essential oil and Yunnan baiyao could lower the level of inflammatory factor TNF-α,IL-6,IL-2 and raise the level of CD4^(+)T cells and CD4^(+)/CD8^(+).Conclusions Tea tree essential oil combined with Yunnan Baiyao in the treatment of experiment oral ulcer can enhance the curative effect,reduce ulcer diameter and inhibit the inflammatory response.
