摘　　要：目的 探究ATG4A蛋白及mRNA在卵巢癌组织中的表达情况及临床意义。方法 收集71例卵巢癌患者作为研究对象,使用Western-blot法检测ATG4A蛋白表达情况,使用RT-PCR技术检测ATG4A mRNA表达水平;分析ATG4A与卵巢癌临床病理参数、化疗效果和患者预后的关系。结果 卵巢癌组织中ATG4A蛋白及ATG4A mRNA表达明显高于癌旁组织(P<0.001)。ATG4A表达水平与患者肿瘤分期、有无淋巴结转移及肿瘤分化程度相关(P<0.05);化疗获得CR、PR的患者癌组织中ATG4A表达量显著低于PD患者(P<0.05);ATG4A高表达患者生存率明显低于低表达患者(P<0.05)。结论 卵巢癌组织中ATG4A表达升高,且与患者肿瘤分期、淋巴结转移、分化程度、患者化疗效果及预后相关,可能可以作为卵巢癌化疗效果及预后的相关评估指标。Objective To investigate the expression of ATG4A protein and mRNA in ovarian cancer tissue and its clinical significance.Methods 71 patients with ovarian cancer were collected as research objects.Western-blot assay was used to detect ATG4A protein expression, and RT-PCR was used to detect ATG4A mRNA expression.To analyze the relationship between ATG4A and clinicopathologic parameters, chemotherapy effect and prognosis of ovarian cancer.Results The expression of ATG4A protein and ATG4A mRNA in ovarian cancer tissues was significantly higher than that in paracancer tissues(P<0.001).The expression level of ATG4A was correlated with tumor stage, lymph node metastasis and tumor differentiation(P<0.05).The expression of ATG4A in cancer tissues of CR and PR patients was significantly lower than that of PD patients(P<0.05).The survival rate of patients with high ATG4A expression was significantly lower than that of patients with low ATG4A expression(P<0.05).Conclusion The increased expression of ATG4A in ovarian cancer tissues is correlated with tumor stage, lymph node metastasis, differentiation degree, chemotherapy effect and prognosis of patients, and may be used as a related indicator of chemotherapy effect and prognosis of ovarian cancer.
