摘　　要：目的:探讨角蛋白17(KRT17)在结肠癌细胞和组织中的表达及生物学功能。方法:免疫组化法、Western blot检测KRT17在结肠癌组织和癌旁组织中的表达,结肠癌细胞分别转染阴性对照组原液KRT17-control和小鼠抗人单克隆抗体anti-KRT17作为对照组和KRT17组。结果:KRT17在结肠癌组织中高表达,KRT17高表达率临床模型诊断结肠癌进展和预后发生的标准误为0.066,AUC(95%CI)为0.847(0.802~0.911),敏感度为71.15%、特异度为82.22%。转染KRT17后,结肠癌SW480细胞增殖能力、迁移率、侵袭能力提高,细胞凋亡率降低。结论:KRT17在结直肠癌细胞和组织中呈高表达,KRT17高表达的结肠癌细胞具有更强的增殖、迁移和侵袭能力,建立KRT17表达的临床模型可预测结肠癌患者的进展和预后。Objective:To investigate expression and biological function of keratin 17(KRT17)in colon cancer cells and tissues.Methods:Expression of KRT17 in colon cancer and adjacent normal mucosa was detected by immunohistochemistry and Western blot.Colon cancer cells were transfected into stock solution of negative control group KRT17-control and mouse anti human monoclonal antibody anti-KRT17 as control group and KRT17 group respectively.Results:KRT17 was highly expressed in colon cancer tissues.Standard error of KRT17 high expression clinical model in diagnosis of progression and prognosis of colon cancer was 0.066,AUC(95%CI)was 0.847(0.802~0.911),sensitivity was 71.15%,specificity was 82.22%.After transfection with KRT17,proliferation,migration and invasion of colon cancer SW480 cells were promoted,and apoptosis rate was decreased.Conclusion:KRT17 is highly expressed in colorectal cancer cells and tissues.Colon cancer cells with high expression of KRT17 have higher proliferation,migration and invasion ability.Establishment of clinical model of KRT17 expression can predict progress and prognosis of colon cancer patients.
