摘  要:目的 探讨牙龈卟啉单胞菌(Porphyromonas gingivalis,P. gingivalis)酪氨酸激酶(Ptk1)致病性的分子机制。方法 采用重组PCR技术构建P. gingivalis野生菌株ATCC 33277的Ptk1单基因缺失的突变菌株(ΔPtk1),通过Real-time PCR技术检测并比较参与调控P. gingivalis(野生型P. gingivalis ATCC 33277与突变型ΔPtk1)细胞外多糖(extracellular polysaccharides,EPS)合成的转录因子SinR的表达情况,同时采用激光共聚焦显微镜观察Ptk1缺失的突变菌株与野生型菌株EPS的形成情况,最后通过ELISA试剂盒检测并比较Ptk1缺失的突变菌株与野生型菌株白细胞介素-1β(IL-1β)表达情况。结果 与野生菌株P. gingivalis ATCC 33277比较,Ptk1单基因缺失的突变菌株转录因子SinR的表达量没有显著变化(t=–1.572,P>0.05);ELISA检测发现,Ptk1单基因缺失的突变菌株IL-1β的表达量较野生型菌株显著下降,差异有统计学意义(t=3.192,P<0.05)。结论 牙龈卟啉单胞菌Ptk1对牙龈卟啉单胞菌细胞外多糖合成和IL-1β的表达有显著调控作用,抑制牙龈卟啉单胞菌酪氨酸激酶的表达或可对植体周围炎的防治有积极作用。Objective To explore the molecular biological mechanism of protein tyrosine kinase(Ptk1) of Porphyromonas gingivalis(P. gingivalis) in the prevention and treatment of peri-implantitis. Methods Recombinant PCR was used to construct single gene deletion mutant(SGDM) strain(ΔPtk1) of wild strain P. gingivalis ATCC 33277 Ptk1. The transcription factor SinR involved in the regulation of extracellular polysaccharide synthesis by P. gingivalis(wild type P. gingivalis ATCC 33277 and mutant ΔPtk1) was detected using Real-time PCR. The expression of extracellular polysaccharides in the two strains were observed with laser confocal microscopy. Finally, the expressions of interleukin-1β in both strains were detected with ELISA assay. Results Compared with the wild strain, the expression level of transcription factor SinR in Ptk1 SGDM strain did not change significantly(t =-1.572,P>0.05). The results of ELISA showed that the expression of IL-1β significantly decreased in the Ptk1 SGDM mutant strain compared with wild strain(t = 3.192, P<0.05).Conclusion P. gingivalis Ptk1 has significant regulatory effect on extracellular polysaccharide synthesis and IL-1β expression in P. gingivalis. Controlling of P. gingivalis Ptk1 may positively contribute to the prevention and treatment of peri implantitis.
