摘  要:研究燕麦蒽酰胺干预对食物过敏模型小鼠血清代谢特征的变化,通过ELISA及UHPLC-QE-MS技术分别对小鼠血清进行测定,经皮致敏加灌胃卵清蛋白使小鼠血清中特异性抗体、过敏介质与细胞因子的含量显著升高,蒽酰胺干预后显著抑制以上免疫因子的释放。OPLS-DA模型显示,空白组(NS)、模型组(OVA)与模型干预组(A-OVA)组间区分明显。食物过敏前后筛选出42个差异代谢物,11条有关氨基酸、糖类等代谢的关键通路;干预后,筛选出54个差异代谢物,6条有关氨基酸、脂肪酸等代谢的关键通路;经食物过敏及蒽酰胺共同影响后,筛选出24个与二者因素相关的差异代谢物。食物过敏及蒽酰胺干预均可对小鼠血清的代谢特征产生显著影响,蒽酰胺对食物过敏具有明显的缓解作用;通路分析显示脂肪酸、氨基酸、维生素B的代谢与食物过敏密切相关。To study the changes of serum metabolic characteristics of food allergy model mice induced by avenanthramides intervention. The serum of mice was determined by ELISA and UHPLC-QE-MS, respectively. Transdermal sensitization and gavage of ovalbumin significantly increased the content of specific antibodies, allergic mediators and cytokines in the serum of mice. Avenanthramides intervention the OPLS-DA model revealed that the blank group(NS), the model group(OVA) and the model intervention group(A-OVA) were significantly distinguished;42 differential metabolites were screened out before and after food allergy, 11 key pathways related to the metabolism of amino acids and carbohydrates;after intervention, 54 differential metabolites and 6 key pathways related to the metabolism of amino acids and fatty acids were screened;after the combined influence of food allergies and anthracene amide, screened out 24 differential metabolites related to the two factors. It was concluded that both food allergy and avenanthramides intervention had significant effects on the metabolic characteristics of mouse serum, and avenanthramides had a significant relieving effect on food allergy. Pathway analysis indicated that the metabolism of fatty acids, amino acids, and vitamin B was closely related to food allergy.
