摘  要:为研究表面活性剂对剩余污泥中噬菌体提取率的影响,利用表面活性剂(焦磷酸钠和柠檬酸钾)提取剩余污泥中的噬菌体,探究噬菌体提取率与污泥胞外聚合物(extracellular polymeric substances, EPS)组分的相关性。在此基础上,利用标记重捕法探究外源T3噬菌体在不同形态EPS中的分布。结果表明:焦磷酸钠和柠檬酸钾分别使剩余污泥中噬菌体提取率提高了283%和79%;噬菌体提取率与剩余污泥EPS溶出的腐殖酸含量有较强相关性,与溶出的多糖和蛋白质含量相关性较弱;利用标记重捕法发现污泥黏附型EPS、松散型EPS和紧密型EPS中外源T3噬菌体回收率分别为69%、15%和34%,表明外源T3噬菌体主要分布于污泥系统的黏附型EPS;EPS中外源T3噬菌体拷贝数与其溶出的腐殖酸含量有较强的相关性,与溶出的多糖和蛋白质含量相关性较弱。To study the effect of surfactants on the extraction rate of phages from waste activated sludge, the phages were extracted from sludge by surfactants(sodium pyrophosphate and potassium citrate), and the correlation between the extraction efficiency of phages and extracellular polymeric substances(EPS) was explored. On this basis, the distribution of exogenous T3phages in EPS with different forms was studied by marking and recaptured method. The results show that sodium pyrophosphate and potassium citrate increase phage extraction efficiency by 283% and 79% in waste activated sludge, respectively. Phage extraction efficiency is strongly correlated with humic acid solubilized from EPS of waste activated sludge, and weakly correlated with polysaccharide, protein solubilized from EPS. Marking and recaptured method finds the recovery of exogenous T3phage are 69%, 15% and 34% in slime EPS, loosely bound EPS and tightly bound EPS, respectively, which indicates that exogenous phages mainly distribute in slime EPS in sludge system;the copy number of exogenous T3phage is strongly correlated with humic acid solubilized from EPS, and weakly correlated with polysaccharide, protein solubilized from EPS.
